Why this applies to Prucalopride

Prucalopride falls under ORAL DRUGS — the group Glucomannan interacts with.

Theoretically, glucomannan may decrease absorption of drugs taken orally.
Due to its viscosity and bulking effects, there is concern that glucomannan can decrease the absorption of oral drugs. A small clinical study in healthy volunteers shows that taking glyburide 2.5 mg plus glucomannan 3.9 grams with breakfast reduces plasma levels of glyburide when compared with breakfast and glyburide alone. In addition, animal research demonstrates this effect on amoxicillin, but shows increased absorption of metronidazole. This mouse model also demonstrates that metronidazole elimination is prolonged, but amoxicillin elimination is enhanced by 38%; glucomannan may also affect the distribution of some drugs. To avoid changes in absorption, take glucomannan 30-60 minutes after taking oral drugs.

Here's something worth knowing about pairing these two. Glucomannan is a thick, gel-forming fiber, and when it's sitting in your stomach and intestines it can physically slow down or trap other oral medicines, meaning less of them actually gets absorbed. Since prucalopride (Motegrity) is swallowed as a pill, there's a reasonable concern the fiber could blunt how much you take in.

If that happens, prucalopride may simply work less well, so your constipation might not improve as much as expected. The good news is this is easy to manage with timing. A simple fix is to take your prucalopride first, then wait about 30 to 60 minutes before taking glucomannan. Always run this past your pharmacist or doctor before changing anything, especially since both are being used for bowel issues.

Glucomannan is a soluble fiber from the konjac root that swells into a gel in your stomach, which may help with mild weight loss, constipation, and modestly lowering cholesterol. What Is Prucalopride Used For?

Brand names: Resotran, Resolor, Motegrity

Prucalopride tablets — sold under the brand name Motegrity and also available as generic prucalopride and prucalopride succinate — are indicated for the treatment of chronic idiopathic constipation (CIC) in adults. CIC is ongoing constipation with no identified underlying cause.

This indication applies to all approved oral tablet forms of prucalopride. It is not approved for use in children; a study in pediatric patients was stopped early because the medication did not show effectiveness in that age group.
