Asafoetida in food amounts appears to be generally well tolerated. At medicinal doses, though, the safety data is thin — we don't have solid information on how safe higher amounts really are.
Large doses have been tied to swelling of the lips, belching, gas, diarrhea, headache, and in rare cases convulsions. Doses of 50–100 mg specifically have caused headache or convulsions in reports.
Asafoetida has also been associated with a serious blood condition (methemoglobinemia) in infants, including one documented case of a 3-month-old given asafoetida for colic who developed dangerously high levels and required intensive care — though he recovered fully. Topically, it's been reported to cause genital swelling.

Theoretically, asafoetida might increase the risk of bleeding when used concomitantly with anticoagulant/antiplatelet drugs. Some anticoagulant/antiplatelet drugs include aspirin, clopidogrel (Plavix), diclofenac (Voltaren, Cataflam, others), ibuprofen (Advil, Motrin, others), naproxen (Anaprox, Naprosyn, others), dalteparin (Fragmin), enoxaparin (Lovenox), heparin, warfarin (Coumadin), and others.
Antihypertensive Drugs
Evidence from animal research suggests that asafoetida has hypotensive effects. Theoretically, concomitant use may increase therapeutic effects of antihypertensive therapy due to hypotensive activity of asafoetida. Some antihypertensive drugs include captopril (Capoten), enalapril (Vasotec), losartan (Cozaar), valsartan (Diovan), diltiazem (Cardizem), Amlodipine (Norvasc), hydrochlorothiazide (HydroDiuril), furosemide (Lasix), and many others.
