glucoZyme contains 9 active ingredients — a blend of digestive enzymes and related compounds. Three are carbohydrate-digesting enzymes: amylase breaks down starches, maltase breaks down malt sugars, and sucrase breaks down table sugar.
Protease digests proteins, lipase digests fats, and cellulase breaks down plant fiber. Lactase helps people digest milk sugar (lactose).
The formula also includes chromium picolinate, a mineral involved in blood sugar control, and cinnamon, a spice that some research has examined for glucose effects. The product is made into capsules using hypromellose, a plant-based capsule material, along with cinnamon bark powder.
Does it work?
Strong evidence
Lactase is effective for lactose intolerance — if you can't digest milk sugar well, this enzyme helps. For the other active ingredients, the evidence we hold doesn't establish clear effectiveness.
Chromium picolinate is likely effective for chromium deficiency and possibly effective for diabetes, but our data shows it as possibly ineffective for prediabetes and other conditions. For protease, lipase, cinnamon, and the remaining enzymes, either no effectiveness ratings are on file or the evidence is insufficient to rate them.

How safe is it?
Well-documented data
Protease is generally well tolerated but may cause digestive upset or allergic reactions. Rare case reports link occupational exposure to airborne proteolytic enzymes with allergic nose and eye symptoms or asthma, though supplement exposure is unlikely to be airborne.
Lipase is generally well tolerated in supplement form but should be used only under medical guidance if you have serious digestive conditions; it has not been studied well in pregnancy or breastfeeding. Lactase is generally well tolerated and likely safe in pregnancy.
A rare case of allergic contact dermatitis to powdered lactase has been reported in a worker. Chromium is usually well tolerated at typical doses but may cause gastrointestinal irritation, headaches, insomnia, irritability, and mood changes; rare serious effects include kidney and liver damage and blood cell problems.
Cinnamon is generally safe in food amounts, but high-dose supplements may harm the liver over time due to coumarin content.

Before taking glucoZyme, double-check with your pharmacist if you take antidiabetes drugs, insulin, or levothyroxine (thyroid medication), since chromium may lower blood sugar or reduce thyroid hormone absorption — both Moderate-severity concerns. NSAIDs and aspirin may increase chromium absorption, though this is a Minor interaction.

Clark Research Association Dr. Clark Zentrum contains two ingredients with documented interactions: chromium picolinate and cinnamon.
The most serious concern is chromium's Moderate-severity interaction with antidiabetes drugs — chromium may lower blood sugar additively, raising the risk of low blood sugar (hypoglycemia), and dose adjustments to your diabetes medications might be needed.

Chromium also has Moderate interactions with levothyroxine (thyroid medication), where it may reduce how much thyroid hormone your body absorbs, and with insulin, which carries the same hypoglycemia risk as other diabetes drugs. NSAIDs and aspirin have Minor interactions with chromium — these pain relievers may increase how much chromium your body absorbs.
Cinnamon carries a Moderate interaction with antidiabetes drugs for the same reason as chromium: it may lower blood sugar on top of what your medication does. It also has a Moderate interaction with drugs that are hard on the liver (hepatotoxic drugs) — large doses of cinnamon contain coumarin, which theoretically could add to that liver burden over time.

Cinnamon
Antidiabetes Drugs
Theoretically, cassia cinnamon may have additive effects with antidiabetes drugs.
Cassia cinnamon may lower blood glucose levels, and have additive effects in patients treated with antidiabetic agents. Dose adjustments to diabetes medications might be necessary.
Hepatotoxic Drugs
Theoretically, large doses of cassia cinnamon might cause additive effects when used with hepatotoxic drugs.
There is some concern that ingesting large amounts of cassia cinnamon for an extended duration might cause hepatotoxicity in some people. Cassia cinnamon contains coumarin, which can cause hepatotoxicity in animal models. In humans, very high doses of coumarin from 50-7000 mg/day can result in hepatotoxicity that resolves when coumarin use is discontinued. Lower amounts might also cause liver problems in sensitive people, such as those with liver disease or those taking potentially hepatotoxic agents.
Chromium Picolinate
Antidiabetes Drugs
Theoretically, chromium may have additive effects with antidiabetic agents and increase the risk of hypoglycemia.
Some research shows that taking chromium might lower blood glucose levels, especially in patients with poorly controlled type 2 diabetes.
Insulin
Theoretically, concomitant use of chromium and insulin might increase the risk of hypoglycemia.
In clinical research, chromium has been shown to increase insulin sensitivity,
Levothyroxine (Synthroid, Others)
Chromium might bind levothyroxine in the intestinal tract and decrease levothyroxine absorption.
Clinical research in healthy volunteers shows that taking chromium picolinate 1000 mcg with levothyroxine 1 mg decreases serum levels of levothyroxine by 17% when compared to taking levothyroxine alone. Advise patients to take levothyroxine at least 30 minutes before or 3-4 hours after taking chromium.
Aspirin
Theoretically, aspirin might increase chromium absorption.
Animal research suggests that aspirin may increase chromium absorption and chromium levels in the blood.
Nonsteroidal Anti-Inflammatory Drugs (Nsaids)
NSAIDs might increase chromium levels in the body.
Drugs that are prostaglandin inhibitors, such as NSAIDs, seem to increase chromium absorption and retention.
