For anxiety, athletic performance, teeth grinding (bruxism), and premenstrual dysphoric disorder (PMDD), we don't have enough reliable evidence to say whether it works or not.
How safe is it?
Well-documented data
L-tryptophan is generally well tolerated by mouth, but common side effects include belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting. A rare serious concern is eosinophilia-myalgia syndrome (EMS), a neurological disorder — but nearly all documented cases in the 1989–1990 outbreak were linked to contaminated L-tryptophan from a single Japanese manufacturer and led to an FDA import alert.
Modern supplements should not carry that contamination risk, though the history warrants caution. During pregnancy, there is not enough safety data, so it's best avoided.
While breastfeeding, safety is not established, so it should be avoided as well. Talk with your doctor or pharmacist about whether L-tryptophan is right for you if you're pregnant or nursing.
