What’s inside
Full disclosure
This supplement contains one active ingredient: L-tryptophan, an amino acid your body uses to produce serotonin and melatonin — both involved in mood, sleep, and nervous-system function. The capsule also holds inactive ingredients (gelatin, magnesium stearate, and rice flour) that serve as the shell and filler.
Does it work?
Not established
The evidence for L-tryptophan in depression is rated possibly ineffective based on available studies. For anxiety, athletic performance, bruxism (teeth grinding), and premenstrual dysphoric disorder, we don't have enough reliable evidence to rate whether it works — the data simply isn't established yet.
How safe is it?
Well-documented data
Orally, L-tryptophan is generally well tolerated, though common side effects include belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting. The supplement does carry a history of product contamination — in 1989, L-tryptophan from a single Japanese manufacturer was linked to over 1,500 cases of a serious neurological disorder called eosinophilia-myalgia syndrome (a condition affecting muscle and blood cells).
Nearly all cases traced back to that one contaminated batch, and the product was recalled and restricted in the U.S. afterward. During pregnancy, safety data is mixed — some evidence suggests it's possibly unsafe, so it's best avoided until you talk with your doctor.
While breastfeeding, safety isn't established, so avoid it and discuss alternatives with your healthcare provider.
Meds to double-check
Major interaction found
Check with your doctor or pharmacist before adding L-tryptophan if you take CNS depressants (drugs like sedatives, sleep aids, or opioid painkillers) — there's a Major interaction risk of extra drowsiness and sedation. Also double-check if you're on serotonergic drugs, especially antidepressants — a Moderate interaction could increase your risk of serotonin syndrome.
The bottom line
Scorecard at a glanceFully disclosed formula with no established evidence rating for its marketed use. Major medication interactions have been identified, and safety information is well characterized.
L-tryptophan may appeal to you if you're exploring natural approaches to mood or sleep, but the evidence for depression is weak and the side-effect profile includes drowsiness and digestive upset. If you take any CNS depressants or serotonergic medications (especially antidepressants), this is a major interaction concern and needs a conversation with your doctor or pharmacist first.
Pregnant or breastfeeding? Skip it and check with your own healthcare provider about what's safe.

L-Tryptophan 500 mg by Best Naturals Drug Interactions
HelloPharmacist Interaction Report
L-Tryptophan 500 mg by Best Naturals interacts with CNS depressants and serotonergic drugs through its amino-acid content.
The most serious concern is with CNS depressants (drugs that slow the brain and nervous system) — combining them with L-tryptophan may cause additive sedative effects, and the supplement itself can trigger fatigue and drowsiness.
Read the full breakdown — every affected drug type, severity by severity
Serotonergic drugs (those that work through serotonin, like certain antidepressants) carry a Moderate interaction risk. L-tryptophan is a building block your body uses to make serotonin, so pairing it with serotonergic medications might amplify serotonin activity in ways that could lead to serotonin syndrome or narrowing of blood vessels in the brain.
Altogether, these interactions span 394 individual medications. Before you start L-tryptophan, use the interaction checker on this page to look up your exact prescriptions, then talk with your doctor or pharmacist about whether it's right for you.
