Lipase and Lactase are generally well tolerated when used as directed. Phytase is considered low-risk in foods, but human supplement data are limited, so talk with your doctor before relying on it.

Protease (papain) is usually tolerated short-term at normal doses but can cause allergic reactions in sensitive people—including itchy eyes, sneezing, or stomach cramps. High doses of papain may be unsafe and could cause severe stomach irritation or esophageal damage.

Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.
