Increased artemether/lumefantrine exposure and an increased risk of QT prolongation
Interaction Deep Dive
Avoid concomitant use of drugs that prolong the QTc or QT interval, such as artemether/lumefantrine as it may add to the QTc or QT effects of entrectinib and increase the risk of cardiac arrhythmias. If concomitant use cannot be avoided, assess QT interval and electrolytes prior to initiation of entrectinib and periodically during treatment. Based on the severity of QTc interval prolongation, withhold entrectinib and then resume at same or reduced dose, or permanently discontinue1. Coadministration of artemether/lumefantrine and entrectinib (CYP3A4 inhibitor) may also increase artemether/lumefantrine exposure. If concomitant administration of artemether/lumefantrine and QT prolonging agent is medically required, monitor the ECG. Additionally, caution is advised when administering drugs that prolong the QT interval after completing artemether/lumefantrine therapy due to the long half-life of lumefantrine (3 to 6 days) 2.
