Acetaminophen, Chlorpheniramine, Dextromethorphan Hydrobromide falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Propolis interacts with.

Theoretically, high doses of propolis might increase blood levels of drugs metabolized by CYP2D6.
In vitro research shows that propolis extract can inhibit CYP2D6. However, animal research shows that propolis extract does not significantly affect CYP2D6 activity when administered to rats at doses up to 250 mg/kg. It is postulated that the constituents of propolis that inhibit CYP2D6 in vitro do not have significant effects in vivo due to low bioavailability and hepatic first-pass effect. This effect has not been reported in humans.

This combo cold and flu product contains dextromethorphan, the cough suppressant, which your liver clears using an enzyme called CYP2D6. In a test tube, propolis can slow that enzyme down. If that happened in your body, dextromethorphan could build up and you might feel more drowsy, dizzy, or jittery than usual. The chlorpheniramine in this product already adds sleepiness, so any extra effect could stack up.

Here's the reassuring part: this is only a theoretical concern from lab studies. When propolis was tested in animals, it didn't meaningfully block CYP2D6, and it has never been reported as a problem in people. Normal propolis use is unlikely to cause trouble, but if you take high-dose supplements, just run it by your pharmacist.

Here's the bottom line on combining propolis with this Coricidin combo product. In a test tube, propolis can slow down a liver enzyme called CYP1A2. The concern is that if that enzyme is blocked, certain ingredients in this cold and flu medicine could build up a bit and hit you harder. For these three pieces, that might mean extra drowsiness or grogginess from the chlorpheniramine (the antihistamine), or more dizziness from the dextromethorphan.

That said, I wouldn't lose sleep over it. This is only a theoretical concern from lab studies. When propolis was actually given to animals, it did not meaningfully change this enzyme, and nothing like this has been reported in people. Normal propolis use is very unlikely to be a problem. Still, if you take high-dose propolis regularly, mention it to your pharmacist.

Furthermore, animal research shows that propolis extract does not significantly affect CYP3A4 activity when administered to rats at doses up to 250 mg/kg. It is postulated that the constituents of propolis that might in inhibit CYP3A4 in vitro do not have significant effects in vivo due to low bioavailability and hepatic first-pass effect. Here's the gist with this combo cold medicine and propolis. One of its ingredients, dextromethorphan (the cough suppressant), is partly broken down by a liver enzyme called CYP3A4. In a test tube, some propolis compounds can slow that enzyme down. If that happened in your body, dextromethorphan could build up a bit, which in theory might mean more drowsiness, dizziness, or feeling foggy. The chlorpheniramine in this product already makes most people sleepy, so anything adding to that is worth noting.

That said, the evidence here is very weak, coming only from lab and animal studies, and it hasn't been seen in real people. Normal amounts of propolis are unlikely to cause trouble. If you take concentrated propolis regularly, just mention it to your pharmacist.

This combination cold and flu product (Coricidin HBP) contains acetaminophen (Tylenol), which is the piece that connects to propolis here. A small part of acetaminophen is handled by a liver enzyme called CYP2E1, and that pathway is the one that creates a harmful, liver-stressing byproduct. In lab dish studies only, propolis appears to slow CYP2E1 down. In theory, blocking that pathway could shift how acetaminophen is broken down.

Here is the honest part: this has only been seen in test tubes, never in actual people, so it is very weak evidence. At normal, label-directed doses you most likely will not notice anything. The bigger acetaminophen rule still applies: watch your total daily dose and avoid heavy alcohol. If you take propolis regularly or have liver concerns, just mention it to your pharmacist.
