Estro-Assured contains six active ingredients: flax, calcium D-glucarate, maitake whole mushroom, lutein, kudzu root extract, and pomegranate fruit extract. Flax is included for its potential effects on cholesterol, blood sugar, and blood pressure; calcium D-glucarate is thought to support the body's natural detoxification; maitake mushroom is used for immune and blood sugar support; lutein is a carotenoid for eye health; kudzu root extract is included for its isoflavone content; and pomegranate fruit extract provides polyphenol antioxidants.
The product is formulated in a vegetable capsule with magnesium stearate as an inactive ingredient.
Does it work?
Not established
The evidence varies by ingredient. Flax is possibly effective for high cholesterol, blood sugar control, high blood pressure, and constipation.
Lutein is possibly effective for age-related macular degeneration and cataracts. Pomegranate is possibly effective for high blood pressure.
For the other uses tested—calcium D-glucarate and maitake mushroom for various cancers, maitake for hay fever and fatigue, kudzu for hay fever and back pain, and pomegranate for cholesterol and diabetes—the evidence we hold is either insufficient to rate or shows the ingredient is possibly ineffective. The data do not establish effectiveness for this product as a whole.
How safe is it?
Well-documented data
Flax is generally well tolerated but commonly causes bloating, gas, and digestive upset, especially at higher doses or without adequate water intake; doses over 45 grams daily may not be tolerated due to diarrhea risk. Maitake mushroom and pomegranate are also generally well tolerated, with mild gastrointestinal effects (diarrhea, nausea, abdominal pain) reported in small numbers of trial participants; one case of rash and itching with maitake has been reported.
Lutein from typical supplement doses is generally well tolerated. Calcium D-glucarate appears well tolerated in small studies, but long-term safety in humans is not established.
Flax is rated possibly unsafe in pregnancy and should be avoided unless approved by your doctor; calcium D-glucarate and maitake mushroom concentrated supplements should be avoided in pregnancy and breastfeeding due to insufficient safety data. Kudzu should be avoided in pregnancy and breastfeeding.
Lutein is likely safe in pregnancy, though supplement doses beyond food amounts warrant caution. For flax and pomegranate, breastfeeding safety data for supplement doses is limited; use caution and discuss with your doctor or pharmacist.

Before taking Estro-Assured, check with your doctor or pharmacist if you take blood thinners (warfarin or other anticoagulants or antiplatelet drugs)—maitake mushroom and pomegranate may increase bleeding risk. Also confirm use if you take blood pressure medications, diabetes drugs, estrogen or tamoxifen, methotrexate, caffeine-containing products, ACE inhibitors, statins (especially rosuvastatin), or drugs metabolized by liver enzymes (CYP2D6, CYP2C9, or CYP3A4 substrates).
Flax may also interact with antibiotics.

The most serious documented interaction is with warfarin (a blood thinner), where both maitake mushroom and pomegranate may increase bleeding risk—maitake through enhanced anticoagulant effects and pomegranate through potential warfarin level increases.

Flax, maitake mushroom, kudzu root extract, and pomegranate all interact with blood pressure medications (antihypertensive drugs), each theoretically lowering blood pressure further and raising the risk of dizziness or fainting. Flax, maitake mushroom, kudzu root extract, and pomegranate also interact with diabetes medications, potentially increasing the risk of low blood sugar (hypoglycemia).
Flax and kudzu may affect blood thinners (anticoagulant and antiplatelet drugs) and raise bleeding risk. Flax interacts with estrogen therapy and antibiotics; kudzu with estrogen therapy, tamoxifen, methotrexate, hepatotoxic drugs, and caffeine; and pomegranate with ACE inhibitor blood pressure drugs and several drugs metabolized by liver enzymes (CYP2D6, CYP2C9, CYP3A4 substrates, and carbamazepine).

Pomegranate Fruit Extract
Ace Inhibitors (Aceis)
Theoretically, taking pomegranate with ACEIs might increase the risk of adverse effects.
Pomegranate juice is thought to have ACE inhibitor-like effects.
Antihypertensive Drugs
Theoretically, taking pomegranate with antihypertensive drugs might increase the risk of hypotension.
Consuming pomegranate juice can modestly lower blood pressure.
Cytochrome P450 2D6 (Cyp2D6) Substrates
Theoretically, pomegranate might increase levels of drugs metabolized by CYP2D6.
In vitro, pomegranate juice inhibits CYP2D6. However, the clinical significance of this potential interaction in humans is not known.
Rosuvastatin (Crestor)
Theoretically, taking pomegranate with rosuvastatin might increase the risk of adverse effects.
In one case, a patient taking rosuvastatin 5 mg every other day in combination with ezetimibe 10 mg daily developed rhabdomyolysis after drinking pomegranate juice 200 mL twice weekly for 3 weeks. This patient had a history of elevated creatine kinase levels while not receiving any statin treatment. This suggests a possible underlying myopathy and predisposition to rhabdomyolysis.
Warfarin (Coumadin)
Theoretically, pomegranate might increase warfarin levels and increase the risk of bleeding. Also, discontinuing regular consumption of pomegranate juice might decrease warfarin levels.
In one case report, a patient had a stable, therapeutic bleeding time, as measured by international normalized ratio (INR), while taking warfarin in combination with pomegranate juice 2-3 times per week. The patient became subtherapeutic within about 10 days after discontinuing pomegranate juice, which required a warfarin dose increase. In another case report, a patient with a stable INR for over one year presented with an INR of 14. The patient noted no changes to medications or diet but did report consuming around 3 liters of pomegranate juice over the previous week. The patient's INR stabilized upon moderation of pomegranate juice consumption. The mechanism of this potential interaction is unclear.
Carbamazepine (Tegretol)
Theoretically, taking pomegranate with carbamazepine might increase the risk of adverse effects, although research suggests this interaction is unlikely to be clinically significant.
Animal research shows that pomegranate juice may inhibit cytochrome P450 3A4 (CYP3A4) metabolism of carbamazepine and increase levels of carbamazepine by 1.5 times without prolonging the elimination half-life. This suggests that pomegranate juice inhibits intestinal CYP3A4, but might not inhibit hepatic CYP3A4. However, some human research suggests that pomegranate does not significantly inhibit CYP3A4 drug metabolism in humans.
Cytochrome P450 2C9 (Cyp2C9) Substrates
Theoretically, pomegranate might increase levels of drugs metabolized by CYP2C9.
Some animal and in vitro research shows that pomegranate juice inhibits intestinal, but not hepatic, CYP2C9 isoenzyme activity. However, clinical research shows that neither pomegranate juice nor pomegranate extract have a significant effect on CYP2C9 activity in humans.
Cytochrome P450 3A4 (Cyp3A4) Substrates
Theoretically, pomegranate might increase levels of drugs metabolized by CYP3A4, but most research suggests this interaction is unlikely to be clinically significant.
Pomegranate contains several polyphenols that have individually been shown to inhibit CYP3A4. However, there is contradictory evidence about the effect of whole pomegranate juice on CYP3A4 activity. In vitro, pomegranate juice significantly inhibits the CYP3A4 enzyme, with comparable inhibition to grapefruit juice. In an animal model, pomegranate juice inhibits CYP3A4 metabolism of carbamazepine and increases levels of carbamazepine by 1.5 times; however, in human volunteers, drinking a single glass of pomegranate juice 240 mL or taking 200 mL daily for 2 weeks does not significantly affect levels of the CYP3A4 substrate midazolam after oral or intravenous administration. Another study in healthy volunteers shows that consuming pomegranate juice 300 mL three times daily for three days also does not significantly affect levels of simvastatin, a CYP3A4 substrate This suggests that pomegranate is unlikely to significantly affect levels of CYP3A4 substrates in humans.
Tolbutamide (Orinase)
Theoretically, pomegranate might increase levels of tolbutamide, although research suggests this interaction is unlikely to be clinically significant.
Animal research shows that pomegranate juice inhibits the cytochrome P450 2C9 (CYP2C9) metabolism of tolbutamide. Pomegranate juice increased tolbutamide levels by 1.2 times without prolonging the elimination half-life. This suggests that pomegranate juice inhibits intestinal CYP2C9, but might not inhibit hepatic CYP2C9. Despite this evidence, clinical research shows that neither pomegranate juice nor pomegranate extract have a significant effect on CYP2C9 activity in humans. This interaction does not appear to be clinically significant in humans.
Flax
Antibiotic Drugs
Theoretically, antibiotics might interfere with the metabolism of flaxseed constituents, which could potentially alter the effects of flaxseed.
Some potential benefits of flaxseed are thought to be due to its lignan content. Secoisolariciresinol diglucoside (SDG), a major lignan precursor, is found in high concentrations in flaxseed. SDG is converted by bacteria in the colon to the lignans enterolactone and enterodiol. Antibiotics alter the flora of the colon, which could theoretically alter the metabolism of flaxseed.
Anticoagulant/Antiplatelet Drugs
Theoretically, using flaxseed in combination with anticoagulant or antiplatelet drugs might have additive effects and increase the risk of bleeding.
Some clinical evidence suggests that the oil contained in flaxseed can decrease platelet aggregation.
Antidiabetes Drugs
Theoretically, flaxseed might have additive effects when used with antidiabetes drugs and increase the risk for hypoglycemia.
Some clinical research suggests that flaxseed can lower blood glucose levels.
Antihypertensive Drugs
Theoretically, flaxseed might have additive effects when used with antihypertensive drugs and increase the risk of hypotension.
Clinical research shows that daily flaxseed consumption, especially for longer than 12 weeks, modestly reduces blood pressure.
Estrogens
Theoretically, taking flaxseed might decrease the effects of estrogens.
Flaxseed contains lignans with mild estrogenic and possible antiestrogenic effects. The lignans seem to compete with circulating endogenous estrogen and might reduce estrogen binding to estrogen receptors, resulting in an anti-estrogen effect. It is unclear if this effect transfers to exogenously administered estrogens.
Kudzu root extract
Anticoagulant/Antiplatelet Drugs
Theoretically, kudzu may increase the risk of bleeding if used with antiplatelet or anticoagulant drugs.
Kudzu isoflavones are reported to have antiplatelet activity.
Caffeine
Theoretically, taking kudzu with caffeine might increase levels of caffeine.
In healthy males injected with the kudzu constituent puerarin, caffeine clearance and metabolism is inhibited. This effect has been attributed to inhibition of cytochrome P450 1A2 (CYP1A2) enzyme, which is involved in caffeine metabolism. It is unclear if taking kudzu orally would have this same effect.
Estrogens
Theoretically, kudzu might alter the effects of estrogen therapy.
Some research suggests that kudzu has estrogenic effects. This may enhance or inhibit the effects of estrogen therapy.
Hepatotoxic Drugs
Theoretically, concomitant use might have additive hepatotoxic effects.
There is some concern that kudzu can adversely affect the liver.
Methotrexate (Trexall, Others)
Theoretically, taking kudzu with methotrexate might increase the risk of methotrexate toxicity.
Preclinical research suggests that kudzu extract greatly reduces the elimination and increases the toxicity of methotrexate. Kudzu might inhibit organic anion transporters (OATs) that are responsible for hepatobiliary and renal excretion of anions, similar to the interaction between methotrexate and non-steroidal anti-inflammatory drugs (NSAIDs).
Tamoxifen (Nolvadex)
Theoretically, kudzu might interfere with tamoxifen activity.
Some research suggests that kudzu may have estrogenic effects.
Antidiabetes Drugs
Theoretically, taking kudzu with antidiabetes drugs might increase the risk of hypoglycemia.
Kudzu might lower blood glucose levels and have additive effects in patients treated with antidiabetic agents. The dose of diabetes medications might need to be adjusted.
Maitake whole Mushroom
Antidiabetes Drugs
Theoretically, combining maitake mushroom with antidiabetes drugs might increase the risk of hypoglycemia.
Clinical research shows that taking maitake mushroom polysaccharide (MMP) can lower blood glucose levels in patients with types 2 diabetes.
Antihypertensive Drugs
Theoretically, combining maitake mushroom with antihypertensive drugs might increase the risk of hypotension.
Animal research shows that maitake mushroom can lower blood pressure.
Warfarin (Coumadin)
There is limited evidence that maitake mushroom may increase the anticoagulant effects of warfarin.
In a case report, a patient previously stabilized on warfarin developed an elevated international normalized ratio (INR) of 5.1 after taking maitake mushroom (Grifron-Pro Maitake D-Fraction) 1 drop/kg daily in three divided doses for one week. The elevated INR resolved after holding warfarin for two days, then reducing the dose by 11%. It is thought that the beta-glucan constituent of maitake mushroom might cause warfarin dissociation from proteins, resulting in increased free warfarin levels and increased warfarin effects.
Calcium D-Glucarate
Alcohol (Ethanol)
Theoretically, concomitant use with alcohol might decrease calcium D-glucarate activity.
There is some evidence that urinary excretion of calcium D-glucarate metabolites increases in people consuming alcohol.
Glucuronidated Drugs
Theoretically, calcium D-glucarate might increase the clearance of drugs that undergo glucuronidation.
The calcium D-glucarate metabolite, D-glucaro-1,4-lactone, inhibits the enzyme beta-glucuronidase, reducing deconjugation of glucuronides in the intestine and thereby reducing reabsorption of the drugs.
Kanamycin
Theoretically, calcium D-glucarate may reduce plasma levels of kanamycin.
Calcium D-glucarate may increase the rate of kanamycin elimination, which may decrease its clinical and adverse effects.
