Not established
The evidence for nicotinamide riboside isn't established yet for most conditions we have data on. For age-related macular degeneration, Alzheimer disease, athletic performance, nonalcoholic fatty liver disease, and heart failure, the research is too limited or conflicting to rate whether it works.

How safe is it?
Well-documented data
Nicotinamide riboside is generally well tolerated in short-term studies, though long-term safety isn't fully known. The most common side effects reported are mild: bloating, muscle pain, nausea, itching, sweating, and temporary changes in bowel habits.
One person in a trial reported leg cramps, and some have noted mild muscle soreness. Gastrointestinal discomfort, diarrhea, and indigestion have been reported with combination products containing nicotinamide riboside, though it's unclear whether nicotinamide riboside alone caused those.
There isn't enough safety data in pregnancy or while breastfeeding, so the safety data advises against use in both situations. Talk with your doctor or pharmacist about whether this is right for you if you're pregnant, planning pregnancy, or nursing.
