Caution is advised with the concomitant use of eluxadoline (an OATP1B1 substrate) and an OATP1B1 inhibitor as this can increase eluxadoline exposure and increase the risk of adverse effects. During drug interaction studies, coadministration of eluxadoline and cyclosporine (an OATP1B1 inhibitor) resulted in eluxadoline AUC and Cmax elevations. As a similar reaction cannot be ruled out with other OATP1B1 inhibitors. Reduce the eluxadoline dose to 75 mg twice daily and monitor for impaired mental or physical abilities if concurrent use is required1.
