Ixazomib Citrate falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Calamus interacts with.

Theoretically, taking calamus with drugs metabolized by CYP3A4 might increase drug levels and potentially increase the risk of adverse effects.
In vitro research shows that calamus extract inhibits CYP3A4 enzyme.

Here's the idea behind this pairing. Your body uses a liver enzyme called CYP3A4 to help clear ixazomib (Ninlaro). In lab studies, calamus appears to slow that enzyme down. If that happened in real life, ixazomib could build up a bit higher than expected, which might mean stronger side effects like nausea, diarrhea, low blood counts, fatigue, or tingling and numbness in the hands and feet.

That said, this is only a theoretical concern based on test-tube research, not human studies, so we don't truly know how big the effect would be. Still, ixazomib is a serious cancer medicine, and calamus carries its own safety worries (the FDA banned it from foods). Please don't start or stop anything on your own. Check with your oncology team or pharmacist first.

Calamus is a swamp plant with a long history in Ayurvedic and traditional Chinese medicine, mostly for digestive and nervous-system complaints. Because of these safety concerns and a lack of solid human evidence, it is generally best avoided.

What Is Ixazomib Used For?

Brand name: Ninlaro

Ninlaro (ixazomib) is used to treat multiple myeloma — a cancer of plasma cells in the bone marrow. It is always used together with two other medicines, lenalidomide and dexamethasone, and is approved for adults who have already tried at least one other treatment.

Ninlaro is not recommended as maintenance therapy or for newly diagnosed multiple myeloma in combination with lenalidomide and dexamethasone outside of controlled clinical trials.
