Lofepramine is a tricyclic antidepressant, and on its own it can already leave some people feeling drowsy, foggy, or a bit slowed down. The concern here is simple overlap: calamus has acted like a mild sedative in animal studies (it seems to nudge up a calming brain chemical called GABA), so pairing it with Lofepramine could in theory add to that sleepy effect and make you feel more tired, woozy, or unsteady on your feet.

I want to be honest with you though, this is a theoretical worry based on animal data, not proven human cases. Calamus carries its own safety baggage too (the FDA banned it from foods over a compound called beta-asarone), so it's one I'd steer clear of anyway. Before combining anything with your antidepressant, please check with your pharmacist or doctor first.

Theoretically, taking calamus with drugs metabolized by CYP2D6 might increase drug levels and potentially increase the risk of adverse effects.
In vitro research shows that calamus extract inhibits CYP2D6 enzyme.

Lofepramine is an antidepressant that your liver clears partly through an enzyme called CYP2D6. Lab studies suggest calamus may slow that enzyme down. If that happens in the body, your lofepramine could build up a bit higher than expected, which might mean stronger side effects like dry mouth, constipation, blurred vision, drowsiness, dizziness on standing, or a faster heartbeat.

I want to be honest with you, though: this concern comes only from test-tube research, not from people, so it's theoretical. On top of that, calamus contains beta-asarone, a compound the FDA banned from foods over safety worries, so it's generally best avoided anyway. Please check with your pharmacist or doctor before combining the two.

Lofepramine is a tricyclic antidepressant, and part of how it works comes from its anticholinergic action, meaning it dampens a nerve signal carried by a chemical called acetylcholine. Calamus appears to push in the opposite direction. In lab studies, it blocks the enzyme that breaks down acetylcholine, so more of that signal hangs around. In theory, that could partly offset Lofepramine's anticholinergic effects.

Honestly, this is a theoretical concern based only on test-tube research, not on people taking these together, so I wouldn't lose sleep over it. The bigger issue is that calamus contains beta-asarone, a compound the FDA banned from foods over safety worries, so it's best avoided regardless. Please loop in your doctor or pharmacist before combining anything with Lofepramine.

Calamus is a swamp plant with a long history in Ayurvedic and traditional Chinese medicine, mostly for digestive and nervous-system complaints. However, it contains beta-asarone, a compound linked to cancer and other toxic effects in animal studies, which is why the U.S. FDA banned it from foods. Because of these safety concerns and a lack of solid human evidence, it is generally best avoided.
