Phenylephrine, Tropicamide falls under ANTICHOLINERGIC DRUGS — the group Calamus interacts with.

Theoretically, concurrent use of anticholinergic drugs and calamus might decrease the effectiveness of the anticholinergic drug.
In vitro evidence shows that calamus can inhibit acetylcholinesterase (AChE).

Mydcombi eye drops combine tropicamide (an anticholinergic that blocks acetylcholine) and phenylephrine to widen your pupil for eye exams. Calamus works in the opposite direction. In lab studies it slows the enzyme that breaks down acetylcholine, so more of that chemical hangs around. In theory, that extra acetylcholine could push back against tropicamide and make your pupil dilate less well or wear off sooner.

Honestly, this is a very weak, theory-only concern. The evidence is only from test-tube and animal work, and these are drops used briefly in the eye, not a daily pill. Since calamus itself carries real safety worries, it's worth mentioning to your eye doctor or pharmacist, but there's no need to be alarmed.

Calamus is a swamp plant with a long history in Ayurvedic and traditional Chinese medicine, mostly for digestive and nervous-system complaints. However, it contains beta-asarone, a compound linked to cancer and other toxic effects in animal studies, which is why the U.S. FDA banned it from foods. Because of these safety concerns and a lack of solid human evidence, it is generally best avoided.
