P-GLYCOPROTEIN SUBSTRATES

Armodafinil falls under P-GLYCOPROTEIN SUBSTRATES — the group Delta-9-tetrahydrocannabinol (thc) interacts with.
Most in vitro research suggests that THC can inhibit P-gp and increase the accumulation of probe compounds by reducing P-gp mediated drug efflux. In vitro studies in kidney cell lines show that a 1-hour exposure to CBD and THC inhibits P-gp. THC may also alter the expression of P-gp, although this effect appears to vary based on duration of exposure. Some in vitro research in lymphoblastoid leukemia cell lines indicates that a 1-hour exposure to cannabinoids does not affect P-gp expression, while a prolonged 72-hour exposure decreases P-gp expression. Other in vitro research in these cell lines shows that a 4-hour exposure to THC and CBD induces P-gp gene expression, while exposure for longer than 4 hours and up to 48 hours does not induce P-gp gene expression.

Here's the gist for combining THC with armodafinil (Nuvigil). THC has shown, mostly in lab dishes and not in people, that it can block a transporter called P-glycoprotein (P-gp). That pump normally helps move certain drugs out of cells. If it gets slowed down, a little more of those drugs could hang around in your body.
Since armodafinil rides along with P-gp, the theory is that THC might nudge its levels up a bit, which could mean more of its usual effects like jitteriness, headache, trouble sleeping, a racing heart, or anxiety. Keep in mind this is weak, theoretical evidence, so I wouldn't lose sleep over it. Just give your pharmacist or doctor a heads-up if you use both, and let them know if you notice any of those feelings.

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for these two together: armodafinil can act as a mild inducer of the CYP3A4 enzyme, which is one of the same pathways your body uses to break down THC. In plain terms, taking armodafinil alongside cannabis or a THC product might speed up how fast you clear the THC, so you could feel less of its effect than you'd expect. It's the THC side of the equation that gets nudged here, not your armodafinil.
I want to be honest about the strength of this one: it's theoretical, based on how the enzymes work rather than solid studies in people, so it may not be noticeable for many folks. Nothing here is dangerous, but if you use both regularly and notice a change, chat with your pharmacist or doctor.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

In vitro research shows that cannabis containing THC might modestly inhibit the activity of CYP3A4 enzymes, which might decrease the metabolism of CYP3A4 substrates. It is unclear if this effect is due to THC, other constituents, or the combination. In vitro research also shows that cannabis extracts containing THC modestly inhibit the CYP3A4 metabolism of testosterone; extracts providing the specific cannabinoids cannabidiol (CBD) or cannabigerol (CBG) alone had stronger inhibitory effects than extracts containing both THC and CBD. Conversely, a crossover clinical study in healthy adults shows that oral THC does not inhibit CYP3A4.

Here's the gist of this one: your body partly clears armodafinil (Nuvigil) using a liver enzyme called CYP3A4. Some lab research suggests THC might slightly slow that enzyme down. In theory, that could let a bit more armodafinil linger in your system, which might nudge up its effects, things like trouble sleeping, headache, jitteriness, a faster heartbeat, or nausea.
That said, I wouldn't lose sleep over this. The evidence is weak (test-tube studies only), and one human study found oral THC did not actually block this enzyme. So any real-world effect is likely small and may not happen at all. Still, since both can affect how alert or wired you feel, it's worth a quick chat with your pharmacist or doctor before combining them.

What Is Armodafinil Used For?

Armodafinil tablets (including the brand Nuvigil) are used to improve wakefulness in adults who have excessive sleepiness linked to one of three conditions: obstructive sleep apnea (OSA), narcolepsy, or shift work disorder (SWD).
In obstructive sleep apnea, armodafinil treats the sleepiness itself — not the underlying airway obstruction. If CPAP (a breathing device worn during sleep) is the right treatment for you, your doctor should try that first before adding armodafinil for sleepiness.
