Why this applies to Glasdegib

Glasdegib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Delta-9-tetrahydrocannabinol (thc) interacts with.

Theoretically, THC may increase the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that cannabis containing THC might modestly inhibit the activity of CYP3A4 enzymes, which might decrease the metabolism of CYP3A4 substrates. It is unclear if this effect is due to THC, other constituents, or the combination. In vitro research also shows that cannabis extracts containing THC modestly inhibit the CYP3A4 metabolism of testosterone; extracts providing the specific cannabinoids cannabidiol (CBD) or cannabigerol (CBG) alone had stronger inhibitory effects than extracts containing both THC and CBD. Conversely, a crossover clinical study in healthy adults shows that oral THC does not inhibit CYP3A4.

Here's the gist with Daurismo (glasdegib) and THC. Your body clears glasdegib using a liver enzyme called CYP3A4. Lab studies hint that THC-containing cannabis might slow that enzyme down a little, which could let glasdegib levels creep up. If that happened, you might notice more of its usual side effects, things like muscle cramps and aches, nausea, tiredness, or changes in taste.

The good news: this is very weak, theoretical evidence. It comes from test-tube work, and one human study actually found oral THC did not block CYP3A4 at all. So in real life this is unlikely to cause trouble. Still, glasdegib is a serious cancer medicine, so please loop in your oncology team or pharmacist before using any THC product.

What Is Glasdegib Used For?

Brand name: Daurismo

Daurismo (glasdegib) is approved to treat newly-diagnosed acute myeloid leukemia (AML) — a fast-growing cancer of the blood and bone marrow. It is used specifically in adults who are 75 years of age or older, or in adults of any age whose other health conditions make intensive induction chemotherapy too risky.

Daurismo is not used alone. It is always given together with low-dose cytarabine, a separate chemotherapy medicine, as part of a 28-day treatment cycle.
