Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Mobocertinib Succinate
Mobocertinib Succinate falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group European Barberry interacts with.
There is very preliminary evidence suggesting that berberine, a constituent of European barberry, might inhibit the CYP3A4 enzyme. Theoretically, European barberry might have a similar effect.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Exkivity (mobocertinib): your body relies on a liver enzyme called CYP3A4 to break this medicine down and keep its levels steady. Berberine, the main active compound in European barberry, may slow that enzyme down. If it does, mobocertinib could build up higher than intended, which may mean more of its known side effects, things like diarrhea, nausea, mouth sores, rash, and importantly heart rhythm changes (a longer QT interval).
I want to be honest about the evidence: this is theoretical, based on how berberine behaves in lab settings, not on real-world cases with this drug. Still, mobocertinib is a serious cancer therapy with real heart and gut risks, so I wouldn't combine it with barberry supplements without checking first. Please talk with your oncologist or pharmacist before adding it.

From our pharmacist’s overview on the Mobocertinib Succinate page
“Mobocertinib (Exkivity) is a targeted therapy for a hard-to-treat lung cancer, which makes dosing precision matter and this list worth real caution. Most of the 146 supplements on file can shift the drug's blood level, because mobocertinib is cleared by CYP3A4, an enzyme many botanicals speed up or slow down.”

About European Barberry
What is European Barberry?
- Digestive upset
- Antimicrobial support
- Blood sugar support
- Cholesterol and metabolic support
- Skin conditions
European barberry is a shrub whose root, bark, and berries contain berberine, a bitter plant alkaloid that has been studied mostly in isolated form. Some early research on berberine is promising for blood sugar and cholesterol, but high-quality evidence for the whole barberry herb is limited. Talk with your pharmacist or doctor before using it, and avoid it during pregnancy and breastfeeding.
About Mobocertinib
What Is Mobocertinib Used For?
Brand name: Exkivity
-
Mobocertinib is used to treat a certain type of non-small cell lung cancer (NSCLC) that cannot be removed by surgery and has spread to other parts of the body either during or after treatment with platinum chemotherapy medications. Mobocertinib is in a class of medications called kinase inhibitors. It works by blocking the action of the abnormal protein that signals cancer cells to multiply. This helps stop or slow the spread of cancer cells and may help shrink tumors.
