Colestipol falls under ORAL DRUGS — the group Glucomannan interacts with.

Theoretically, glucomannan may decrease absorption of drugs taken orally.
Due to its viscosity and bulking effects, there is concern that glucomannan can decrease the absorption of oral drugs. A small clinical study in healthy volunteers shows that taking glyburide 2.5 mg plus glucomannan 3.9 grams with breakfast reduces plasma levels of glyburide when compared with breakfast and glyburide alone. In addition, animal research demonstrates this effect on amoxicillin, but shows increased absorption of metronidazole. This mouse model also demonstrates that metronidazole elimination is prolonged, but amoxicillin elimination is enhanced by 38%; glucomannan may also affect the distribution of some drugs. To avoid changes in absorption, take glucomannan 30-60 minutes after taking oral drugs.

Here's something interesting about pairing these two. Glucomannan is a thick, gel-forming fiber, and Colestipol works right in your intestines by grabbing onto bile acids to lower your cholesterol. When you take a bulky fiber like glucomannan around the same time, that gel can physically get in the way and blunt how well other oral medicines are absorbed. Since both of these act in the gut, mixing them too closely could leave you with weaker cholesterol-lowering results.

The good news is the fix is simple timing. To play it safe, separate them, generally take glucomannan about 30 to 60 minutes after your Colestipol. Before you start glucomannan, run it by your pharmacist or doctor so they can confirm the spacing that fits your routine.

Colestipol — available as oral suspension and tablets under brands including Colestid, Flavored Colestid, and Colestipol Hydrochloride — is used as part of a broader treatment plan (including diet) to lower elevated LDL cholesterol in adults with primary hypercholesterolemia (a condition where LDL cholesterol stays high despite dietary changes).

It is not a standalone fix. Diet, weight management, and other lifestyle changes come first, and colestipol is added when those steps alone aren't enough to reach target LDL levels.
