Desvenlafaxine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Greater Celandine interacts with.
Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and many others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with desvenlafaxine (Pristiq) and greater celandine. One compound in the herb, chelidonine, can block a liver enzyme called CYP2D6 in test-tube studies. Desvenlafaxine is partly handled by that pathway, and it is the active drug itself (not a prodrug), so slowing that enzyme could let levels creep up a bit. In real life that might mean more of the usual SNRI side effects, such as nausea, sweating, a faster heartbeat, jitteriness, trouble sleeping, or higher blood pressure.
That said, this is only theoretical lab data, not proven in people, so I wouldn't lose sleep over it. The bigger worry is that greater celandine has been tied to serious liver injury, which is reason enough to skip it. Please check with your pharmacist or doctor before combining them.
