Chlorpheniramine, Phenylephrine, Phenylpropanolamine, Phenyltoloxamine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Guggul interacts with.
In vitro research shows that guggul constituents known as guggulsterones can induce CYP3A4.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
This combination product mixes an antihistamine (chlorpheniramine), a decongestant (phenylephrine), and other cold-and-allergy ingredients to ease sneezing, runny nose, and stuffiness. Some of these get processed by a liver enzyme called CYP3A4. In lab and animal studies, the active compounds in guggul (guggulsterones) appear to speed up that enzyme. If that happened in people, your body might clear parts of this medicine a bit faster, so you could notice less relief, your nose stays stuffy or your allergy symptoms aren't as controlled.
Keep in mind this is theoretical and based only on test-tube and animal work, so it may not amount to much in real life. None of these ingredients are prodrugs that need activating. Still, if you take guggul regularly and feel your cold medicine isn't working as well, mention it to your pharmacist or doctor.
