Theoretically, papaya extract may increase the levels and clinical effects of amiodarone.
Animal research in rats shows that a single oral dose of papaya extract, as well as multiple doses of papaya extract daily over 14 days, prior to a single dose of amiodarone delays the time to maximum amiodarone concentration. However, only the 14-day papaya extract regimen increases systemic amiodarone exposure by 60% to 70%. This interaction has not been reported in humans.

Here's the gist for amiodarone: this is a powerful, long-acting heart rhythm drug with a famously narrow margin between a helpful dose and too much. The concern is that concentrated papaya extract might slow how your body clears amiodarone, letting more of it build up over time. If levels climb, you could see more of amiodarone's typical effects, things like a slower heart rate, dizziness, fatigue, or worsening of its known side effects.

I want to be honest about the evidence though. This comes only from rat studies, where two weeks of papaya extract raised amiodarone exposure by about 60 to 70 percent. It has never been reported in people, and eating ordinary papaya as food is unlikely to be the issue. Concentrated supplements are the real question. Before adding any papaya supplement, please run it by your pharmacist or doctor.

Papaya leaf extract is being studied for raising platelet counts in dengue fever, but most other supplement uses have limited human evidence. What Is Amiodarone Used For?

Amiodarone is prescribed for adults with serious, life-threatening heart-rhythm problems that haven't responded to other medications. Specifically, it's used for two dangerous arrhythmias (abnormal heartbeats): recurrent ventricular fibrillation (a chaotic, potentially fatal heart rhythm) and recurrent ventricular tachycardia (a dangerously fast heart rhythm that causes unstable blood pressure).

Because amiodarone comes with significant risks, doctors reserve it for situations where other options haven't worked or can't be tolerated. It is not a first-choice medication — it's a last-resort tool for serious, life-threatening conditions.
