Why this applies to Glyburide
Glyburide falls under ANTIDIABETES DRUGS — the group Papaya interacts with.
A small low-quality clinical study in patients with type 2 diabetes who are taking glibenclamide shows that taking a fermented papaya preparation 3 grams daily for 2 months decreases fasting and postprandial blood glucose levels when compared to baseline. Additionally, of the 25 patients in the study, 9 required a reduction in glibenclamide dose.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
Here's the gist for glyburide: it lowers your blood sugar by nudging your pancreas to release more insulin. In one small, lower-quality study, people taking a fermented papaya product (3 grams a day for two months) saw their blood sugar drop further, and about a third of them actually needed a lower glyburide dose. So papaya seems to add to glyburide's sugar-lowering punch rather than block it.
The concern is stacking two things that both push blood sugar down, which can tip you into hypoglycemia: shakiness, sweating, dizziness, confusion, or feeling faint. Keep in mind this was studied with a concentrated fermented preparation, not a slice of papaya at breakfast, and we don't know if regular fruit acts the same way. If you want to use a papaya supplement, check with your pharmacist or doctor first and watch your readings closely.

Papaya is a tropical fruit that is nutritious and generally safe to eat as food, and it contains an enzyme called papain used as a digestive aid and meat tenderizer. Papaya leaf extract is being studied for raising platelet counts in dengue fever, but most other supplement uses have limited human evidence. Talk to your pharmacist or doctor before using concentrated papaya supplements, and avoid unripe (green) papaya and papaya seed products in pregnancy.

What Is Glyburide Used For?

Glyburide tablets are used alongside diet and exercise to help control blood sugar in adults with type 2 diabetes. It is not for type 1 diabetes or diabetic ketoacidosis (a serious complication where the body produces dangerous levels of acids called ketones).
Glyburide may be used alone or combined with metformin when metformin alone is not enough. It can also be used to help transition patients who are moving from insulin or other diabetes pills to a new regimen, under a doctor's guidance.
