CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Ethinyl Estradiol, Ethynodiol Diacetate falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Peppermint interacts with.
Clinical research in healthy volunteers shows that a single dose of peppermint oil 600 mg inhibits CYP3A4 enzymes and increases the AUC of felodipine, a CYP3A4 substrate. However, in vitro research suggests that peppermint oil only inhibits CYP3A4 at very high concentrations.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Your birth control pill contains ethinyl estradiol, which your body clears partly through a liver enzyme called CYP3A4. Peppermint oil may slow that enzyme down a little, so in theory the estrogen could build up slightly. If that happens, you might notice more of the usual estrogen-related effects, like nausea, breast tenderness, headaches, or some bloating.
Here is the reassuring part: this is mostly theoretical, and the human study used a concentrated 600 mg dose of peppermint oil. A peppermint tea, a candy, or a bit of mint in food is very unlikely to cause any real change. If you regularly take concentrated peppermint oil capsules (often for IBS), it is worth a quick chat with your pharmacist or doctor.

CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES

In vitro and animal research shows that peppermint oil and peppermint leaf inhibit CYP1A2. However, in clinical research, peppermint tea did not significantly affect the metabolism of caffeine, a CYP1A2 substrate. It is possible that the 6-day duration of treatment may have been too short to identify a difference.

Here's the situation with your birth control pill (Demulen, Zovia, Kelnor) and peppermint. The estrogen part, ethinyl estradiol, is partly handled by a liver enzyme called CYP1A2. In lab and animal studies, peppermint oil seems to slow that enzyme down a little. In theory, that could nudge estrogen levels up slightly, which might mean a touch more of the usual estrogen-related effects like nausea, breast tenderness, or bloating.
The good news: this is rated as a minor, theoretical concern. When researchers actually tested peppermint tea in people, it didn't meaningfully change how the body cleared a similar enzyme target. A cup of tea or a mint is very unlikely to matter. If you plan to use concentrated peppermint oil capsules daily, just mention it to your pharmacist or doctor so they can keep an eye on things.
