Why this applies to Palovarotene

Palovarotene falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Propolis interacts with.

Theoretically, high doses of propolis might increase blood levels of drugs metabolized by CYP3A4.
Some in vitro research shows that propolis extract can inhibit CYP3A4; however, other in vitro research shows that propolis has no effect on CYP3A4 activity. Furthermore, animal research shows that propolis extract does not significantly affect CYP3A4 activity when administered to rats at doses up to 250 mg/kg. It is postulated that the constituents of propolis that might in inhibit CYP3A4 in vitro do not have significant effects in vivo due to low bioavailability and hepatic first-pass effect. This effect has not been reported in humans.

Here's the gist for anyone taking Sohonos (palovarotene) alongside propolis. Palovarotene is a retinoid, meaning it works like a form of vitamin A, and your liver clears it using an enzyme called CYP3A4. Some lab-dish studies hint that propolis might slow that enzyme down. If that happened in the body, palovarotene levels could creep up, which for a retinoid could mean more dry skin and lips, joint aches, headaches, or other vitamin A type side effects.

The good news is that this concern is very weak. Other lab studies showed no effect, and animal research found no real change. It has never been reported in people. Everyday propolis use is unlikely to cause trouble, but since palovarotene is a serious medicine, run any propolis product by your pharmacist or doctor first.

What Is Palovarotene Used For?

Brand name: Sohonos

Sohonos (palovarotene) is used to reduce the volume of new heterotopic ossification — abnormal bone that forms in muscles and soft tissue — in people with fibrodysplasia ossificans progressiva (FOP). It is approved for adults and for children aged 8 years and older (females) and 10 years and older (males).

FOP is a rare genetic condition where the body progressively forms extra bone outside the skeleton, especially after injury or flare-ups. Sohonos is taken both as a daily maintenance dose and at higher doses when a flare-up occurs.
