Lumateperone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Syrian Rue interacts with.

Theoretically, Syrian rue might increase the levels and clinical effects of drugs metabolized by CYP3A4.
Syrian rue contains beta-carboline alkaloids, including harmine, harmol, and harmane. In vitro research suggests that these beta-carboline alkaloids competitively inhibit CYP3A4 enzymes.

Here's the gist for Caplyta (lumateperone): your liver clears it mainly through an enzyme called CYP3A4. Syrian Rue contains harmala alkaloids that, in lab studies, can slow that same enzyme down. If CYP3A4 works more slowly, lumateperone may hang around in your body longer than expected and build up.

For you, that could mean stronger side effects like drowsiness, dizziness, dry mouth, or feeling unsteady. The good news is this concern comes from test-tube research only, not from real people, so it's theoretical at this point. Still, Syrian Rue is potent and not recommended for self-use. Please loop in your pharmacist or doctor before combining the two so they can keep you safe.

What Is Lumateperone Used For?

Brand name: Caplyta

Caplyta (lumateperone) is approved for adults with schizophrenia, depressive episodes associated with bipolar I or II disorder (bipolar depression), and as an add-on treatment for major depressive disorder (MDD).

For bipolar depression, Caplyta can be used on its own or alongside lithium or valproate. For MDD, Caplyta is used together with an antidepressant — not on its own as a standalone antidepressant.
