Why this applies to Adagrasib

Adagrasib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Valerian interacts with.

Valerian does not seem to have a clinically relevant effect on levels of drugs metabolized by CYP3A4.
Although some in vitro evidence suggests that valerian extract might inhibit or induce CYP3A4, clinical pharmacokinetic (PK) studies show that valerian does not have a clinically significant effect on the CYP3A4 enzyme. In one PK study, taking valerian 125 mg three times daily for 28 days did not affect metabolism of midazolam, an accepted CYP3A4 probe-substrate. In another PK study, taking valerian 1000 mg (providing about 11 mg valerenic acid) nightly for 14 days modestly increases levels of alprazolam, a CYP3A4 substrate, suggesting mild inhibition of CYP3A4. However, this mild inhibition is unlikely to be clinically relevant.

If you're taking Adagrasib (Krazati) for cancer and wondering about valerian for sleep, here's the good news. Adagrasib is cleared from your body by an enzyme called CYP3A4, and valerian touches that same enzyme only very slightly. In real-world studies, valerian barely budged the levels of drugs handled by CYP3A4, so it's not expected to meaningfully change how much adagrasib is in your system.

In plain terms, valerian is unlikely to make adagrasib's side effects (like nausea, diarrhea, or fatigue) noticeably worse. This is a minor, mostly theoretical concern. Still, because adagrasib is a serious cancer medicine, please run any new supplement past your oncology pharmacist or doctor first.

About Adagrasib

What Is Adagrasib Used For?

Brand name: Krazati

Krazati (adagrasib) is used to treat adults with locally advanced or metastatic non-small cell lung cancer (NSCLC) that carries a specific genetic change called a KRAS G12C mutation. It is used as a single agent for NSCLC after at least one prior treatment, and patients must be confirmed to have this mutation by an FDA-approved test.

Krazati is also used in combination with another drug called cetuximab to treat adults with KRAS G12C-mutated locally advanced or metastatic colorectal cancer (CRC) who have already been treated with specific prior chemotherapy regimens. Both uses were approved under the FDA's accelerated approval program based on tumor response data, and continued approval may depend on results from confirmatory trials.
