Macitentan falls under HEPATOTOXIC DRUGS — the group Vitamin A interacts with.

Theoretically, taking high doses of vitamin A in combination with other potentially hepatotoxic drugs might increase the risk of liver disease.
The tolerable upper intake level (UL) is the highest level of intake that is likely to pose no risk of adverse effects. Doses of vitamin A above the UL can cause hepatotoxicity, ranging from elevated liver enzymes to liver failure.

Here's the thing to know about pairing Opsumit (macitentan) with vitamin A. This isn't about vitamin A making the drug stronger or weaker. Instead, both can be hard on the liver, and the concern is that stacking them might add up. Macitentan can occasionally raise liver enzymes on its own, and large amounts of vitamin A can do the same, all the way up to more serious liver trouble at very high doses.

To be fair, this is mostly a theoretical worry based on expert opinion, not strong studies. Normal food and a regular multivitamin are very unlikely to be a problem. The real risk comes from big, concentrated vitamin A doses above the safe upper limit. Watch for yellowing skin or eyes, dark urine, nausea, or belly pain, and check with your pharmacist or doctor before adding a high-dose supplement.

What Is Macitentan Used For?

Brand name: Opsumit

Macitentan tablets — sold under the brand name Opsumit — are used to treat pulmonary arterial hypertension (PAH, WHO Group I), a serious condition where the blood pressure in the arteries supplying the lungs is abnormally high. The goal is to slow down how quickly the disease gets worse and to lower the chances of being hospitalized because of it.

In clinical studies, it was shown to be effective in adults with mostly moderate-to-severe PAH symptoms. Patients in those studies had PAH caused by unknown or inherited reasons, connective tissue disorders, or congenital heart disease with repaired heart shunts.
