Diazepam falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Vitamin E interacts with.

Theoretically, vitamin E might induce metabolism of CYP3A4, possibly reducing the levels CYP3A4 substrates.
Vitamin E appears to bind with the nuclear receptor, pregnane X receptor (PXR), which results in increased expression of CYP3A4. Although the clinical significance of this is not known, use caution when considering concomitant use of vitamin E and other drugs affected by these enzymes.

Here's the gist for Valium (diazepam): your liver uses an enzyme called CYP3A4 to help break it down. There's a theory that high-dose vitamin E may gently nudge that enzyme to work a little faster. If that happens, your body could clear diazepam a bit quicker, which might make it feel less effective at calming anxiety, easing muscle spasms, or controlling seizures.

Now, please don't lose sleep over this. This is a theoretical concern based on how vitamin E behaves in lab studies, not something proven in real patients. Everyday food amounts of vitamin E are not the worry; concentrated high-dose supplements are. If you take diazepam regularly and want to add vitamin E, just run it by your pharmacist or doctor first.

What Is Diazepam Used For?

  • Diazepam is used to relieve anxiety and to control agitation caused by alcohol withdrawal. It is also used along with other medications to control muscle spasms and spasticity caused by certain neurological disorders such as cerebral palsy (condition that causes difficulty with movement and balance), paraplegia (inability to move parts of the body), athetosis (abnormal muscle contractions), and stiff-man syndrome (a rare disorder with muscle rigidity and stiffness). Diazepam is also used along with other medications to control seizures. Diazepam is in a class of medications called benzodiazepines. It works by calming abnormal overactivity in the brain.
