Cyclobenzaprine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Vitamin E interacts with.

Theoretically, vitamin E might induce metabolism of CYP3A4, possibly reducing the levels CYP3A4 substrates.
Vitamin E appears to bind with the nuclear receptor, pregnane X receptor (PXR), which results in increased expression of CYP3A4. Although the clinical significance of this is not known, use caution when considering concomitant use of vitamin E and other drugs affected by these enzymes.

Here's the gist for these two. Your body uses an enzyme called CYP3A4 to help break down cyclobenzaprine (Flexeril). The theory is that high-dose vitamin E may gently nudge that enzyme to work a little faster. If that happened, your body could clear the muscle relaxant a bit quicker, which might make it feel less effective at easing that stiff, sore back or neck. Cyclobenzaprine is the active drug itself, not a prodrug, so faster breakdown means a weaker effect, not stronger.

The good news: this is only a theoretical concern based on how the chemicals behave, not real patient reports, and everyday food amounts of vitamin E won't matter. If you're taking high-dose supplements, just mention it to your pharmacist or doctor.

What Is Cyclobenzaprine Used For?

Brand names: Cyclobenzaprine, Flexeril, Cycloprine

Cyclobenzaprine tablets and extended-release capsules are used alongside rest and physical therapy to relieve muscle spasm caused by acute, painful musculoskeletal conditions — easing pain, tenderness, and limited movement. This use is intended for short-term treatment only (up to 2–3 weeks).

Tonmya, the sublingual (under-the-tongue) form of cyclobenzaprine, is separately indicated for the treatment of fibromyalgia in adults. It is not the same as the oral tablet or capsule formulations.
