Does it work?
Insufficient evidence
The evidence for these amino acids varies widely. L-Phenylalanine shows possibly effective evidence for vitiligo but possibly ineffective for ADHD; evidence for alcohol use disorder and multiple sclerosis is insufficient.
L-Lysine is possibly effective for cold sores. L-Histidine, L-Threonine, and the other amino acids either lack sufficient evidence or show inconclusive results in the data we hold.
How safe is it?
Well-documented data
These amino acids are generally well tolerated at typical doses. L-Phenylalanine can cause anxiety, constipation, headache, heartburn, insomnia, nausea, or sedation in some people, and should not be used by anyone with PKU (a rare genetic disorder).
L-Threonine has been associated with headache and minor digestive upset like diarrhea or gas. L-Lysine is generally well tolerated but may cause stomach pain or diarrhea at high doses.
For pregnancy and breastfeeding, the data is limited. L-Phenylalanine should be avoided in supplement doses during pregnancy and breastfeeding.
L-Histidine is likely safe in pregnancy and lactation. For L-Threonine and L-Lysine, supplement safety during pregnancy and breastfeeding has not been well studied—talk with your doctor or pharmacist before use if you are pregnant or nursing.

L-Phenylalanine is the ingredient of most concern. It can significantly reduce how well levodopa (a Parkinson's medication) works by competing with it for transport into the brain—this is a Major interaction that may worsen tremor, stiffness, and the "on-off" effect.
It also moderately reduces absorption of baclofen (a muscle relaxant) and poses a theoretical risk of dangerous blood pressure spikes (hypertensive crisis) if combined with older-type antidepressants called MAOIs.
L-Threonine has a documented Major interaction with NMDA antagonists (a class of brain-acting drugs), theoretically decreasing their effectiveness. L-Lysine carries a Minor interaction with 5-HT4 agonists, which may reduce their effect.
