Flexible Joint Formula contains five active ingredients. Glucosamine sulfate and chondroitin sulfate are the main structural components—they're thought to support joint cartilage and cushioning.
Hyaluronic acid acts as a lubricant in joints and tissues. Cissus quadrangularis extract is a plant-derived ingredient included for joint and bone support.
Methylsulfonylmethane (MSM) is a sulfur-containing compound often added to joint formulas, though we hold limited interaction data for it. The capsules also contain inactive ingredients—wheat flour, titanium dioxide, and gelatin—which serve as fillers and the capsule shell itself.

Glucosamine sulfate is likely effective for osteoarthritis, the most common joint condition. Chondroitin sulfate is possibly effective for osteoarthritis and cataracts.
Cissus quadrangularis extract is possibly effective for obesity. Hyaluronic acid is possibly effective for dry eye and venous leg ulcers (slow-healing sores on the legs from poor circulation).
For other uses listed on the label—aging skin, back pain, and colorectal health—the evidence we hold is insufficient to rate these ingredients reliably, so we can't say whether they work for those purposes.
How safe is it?
Well-documented data
Glucosamine and chondroitin are generally well tolerated when taken by mouth, though common side effects include bloating, constipation, cramps, diarrhea, heartburn, and nausea. Rarely, both have been linked to allergic reactions—about 72% of reported adverse events for glucosamine were hypersensitivity reactions, ranging from mild (itching, hives, lip swelling) to severe (breathing difficulty, dizziness, low blood pressure), and anaphylaxis was reported in 1.5% of cases.
There have also been rare reports of asthma flare-ups with glucosamine–chondroitin combinations. Cissus quadrangularis may cause headache, insomnia, gas, diarrhea, stomach inflammation, or dry mouth, though these occurred at rates similar to placebo in trials.
Hyaluronic acid appears well tolerated orally and topically. Pregnancy and breastfeeding safety data are insufficient for all main ingredients—glucosamine, chondroitin, cissus, and hyaluronic acid (when taken orally as a supplement).

If you use antidiabetes medications, watch for low blood sugar (hypoglycemia), especially with cissus quadrangularis. Avoid combining acetaminophen (Tylenol) with this formula if you're relying on it for pain relief, as glucosamine may reduce its effectiveness.

Both glucosamine and cissus quadrangularis extract carry minor to moderate interactions with antidiabetes drugs: glucosamine is unlikely to interfere much, but cissus might raise the risk of low blood sugar (hypoglycemia) when combined with diabetes medication.
Glucosamine may also reduce pain relief from acetaminophen (Tylenol) in some people with osteoarthritis.

Glucosamine Sulfate
Warfarin (Coumadin)
Glucosamine might increase the anticoagulant effects of warfarin and increase the risk of bruising and bleeding.
In two individual case reports, glucosamine/chondroitin combinations were associated with a significant increase in international normalized ratio (INR) in patients previously stabilized on warfarin. In one case, the increase in INR occurred only after tripling the dose of a glucosamine/chondroitin supplement from 500 mg/400 mg daily to 1500/1200 mg daily. Additionally, 20 voluntary case reports to the U.S. Food & Drug Administration (FDA) have linked glucosamine plus chondroitin with increased INR, bruising, and bleeding in patients who were also taking warfarin. There have also been 20 additional case reports to the World Health Organization (WHO) that link glucosamine alone to increased INR in patients taking warfarin. The mechanism of this interaction is unclear. Glucosamine is a small component of heparin, but is not thought to have anticoagulant activity; however, animal research suggests that it might have antiplatelet activity.
Topoisomerase Ii Inhibitors
Theoretically glucosamine may induce resistance to topoisomerase II inhibitors.
In vitro research suggests that glucosamine might induce resistance to etoposide (VP16, VePesid) and doxorubicin (Adriamycin) by reducing inhibition of topoisomerase II, an enzyme required for DNA replication in tumor cells. This effect has not been reported in humans.
Acetaminophen (Tylenol, Others)
Acetaminophen might interfere with the activity of glucosamine sulfate by interacting with the sulfate portion.
Anecdotal reports suggest that adding glucosamine to an acetaminophen regimen might decrease pain control in patients with osteoarthritis. Some research suggests that the sulfate portion of glucosamine sulfate might contribute to its effect in osteoarthritis. Since acetaminophen metabolism requires sulfur and reduces serum sulfate concentrations, acetaminophen could theoretically interfere with the action of glucosamine sulfate. Conversely, the administration of sulfate could theoretically decrease the effectiveness of acetaminophen in sulfate-deficient people by increasing its clearance.
Antidiabetes Drugs
Despite initial concerns, it is unlikely that glucosamine will interfere with the effects of antidiabetes drugs.
In vitro and animal research has suggested that glucosamine might increase insulin resistance or decrease insulin production. This has raised concerns that taking glucosamine might worsen diabetes and decrease the effectiveness of diabetes drugs. However, clinical research suggests that glucosamine does not have adverse effects on blood glucose or glycated hemoglobin (HbA1C) in healthy, obese, or type 2 diabetes patients.
Cissus quadrangularis extract
Antidiabetes Drugs
Theoretically, Cissus quadrangularis might increase the risk of hypoglycemia when taken with antidiabetes drugs.
Small clinical studies suggest that Cissus quadrangularis might reduce fasting blood glucose in individuals with overweight or obesity.
Chondroitin Sulfate
Warfarin (Coumadin)
Taking chondroitin in combination with glucosamine might increase the anticoagulant effects of warfarin. However, the effect of chondroitin alone is unclear.
There have been multiple reports of increased international normalized ratio (INR) in patients taking warfarin with glucosamine, with or without chondroitin. The lack of reports with chondroitin alone seem to suggest that the interactions occurring in these reports may have been due to glucosamine. In two individual case reports, glucosamine/chondroitin combinations were associated with a significant increase in INR in patients previously stabilized on warfarin. Additionally, 20 voluntary case reports to the US Food & Drug Administration (FDA) have linked glucosamine plus chondroitin with increased INR, bruising, and bleeding in patients who were also taking warfarin.
