The remaining ingredients—dicalcium phosphate, riboflavin, microcrystalline cellulose, croscarmellose sodium, stearic acid, magnesium stearate, silicon dioxide, HPMC, and titanium dioxide—are inactive excipients that bind and preserve the tablet.
Does it work?
Strong evidence
SAMe is likely effective for osteoarthritis, meaning there's solid evidence it can help with joint pain and function. For depression, the evidence rates it as possibly effective—there's some promising research, but we'd need stronger data to be certain.
For liver disease (cholestasis), SAMe is also possibly effective. For Alzheimer disease and ADHD, there isn't enough reliable evidence to know whether it works.
How safe is it?
Well-documented data
SAMe is generally well tolerated at typical doses, but stomach upset is common and can include nausea, diarrhea, constipation, dizziness, and headache. Higher doses raise the risk of side effects.
There's one caution that stands out: SAMe may trigger mania in people with bipolar disorder, so if you have that condition, talk with your doctor before taking it. Safety data overall isn't well established, and you should use this supplement only under the guidance of a healthcare provider.
During pregnancy, SAMe rates as possibly safe, though data is limited; for breastfeeding, there isn't enough information on file to know either way, so discuss it with your doctor or pharmacist.

Formula
S-Adenosyl Methionine (SAMe) is derived from the amino acid methionine and is one of the most important methyl donors in the central nervous system. Methylation (donation of methyl groups) is an essential process in ensuring the proper function of a number of organ systems. SAMe is involved in numerous biochemical reactions in tissues, including the liver, joints and brain. SAMe is also required for the biosynthesis of critical neurotransmitters and hormones.

Levodopa
SAMe might reduce the effectiveness of levodopa.
SAMe methylates levodopa, which might reduce its effectiveness for treating Parkinson disease.
Serotonergic Drugs
Taking SAMe with serotonergic drugs might increase the risk of serotonin syndrome and other serotonergic side effects.
SAMe has serotonergic effects. Theoretically, combining serotonergic drugs with SAMe might increase the risk of serotonergic side effects, including serotonin syndrome and cerebral vasoconstrictive disorders. In one case report, SAMe 100 mg intramuscularly was given daily with clomipramine (Anafranil) 25 mg per day. When the clomipramine dose was increased to 75 mg per day the patient experienced serotonin syndrome about 48-72 hours later, requiring hospitalization.
