Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Acalabrutinib
Acalabrutinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group African Wild Potato interacts with.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for Calquence (acalabrutinib). Your liver uses an enzyme called CYP3A4 to break this medicine down. In lab dish studies, African Wild Potato slowed that enzyme quite a bit. If the same thing happened in your body, acalabrutinib could clear more slowly and build up, which might mean stronger side effects like bruising or bleeding, headaches, low blood counts, infections, or an irregular heartbeat.
Now, the important part: this concern comes only from laboratory work, and in one human study the supplement didn't actually change how a similar CYP3A4 drug behaved. So this is more of a theoretical caution than a proven problem. Still, with a cancer therapy like this, I'd rather you not gamble. Please check with your oncologist or pharmacist before adding this supplement.

From our pharmacist’s overview on the Acalabrutinib page
“For anyone taking acalabrutinib (Calquence) for chronic lymphocytic leukemia or lymphoma, the supplement list below deserves genuine scrutiny. Of 144 ingredients on file, nearly all act on CYP3A4, the liver enzyme that clears the drug, or on P-glycoprotein, a transporter that moves it, so they can push its level too high or leave too l…”
