Why this applies to Capmatinib Hydrochloride

Capmatinib Hydrochloride falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group African Wild Potato interacts with.

In vitro evidence shows that African wild potato extract can inhibit cytochrome P450 3A4 (CYP3A4) enzyme activity by up to 86%. Theoretically, African wild potato might increase levels of drugs metabolized by CYP3A4. However, in human research, African wild potato does not seem to affect the pharmacokinetics of Efavirenz, a CYP3A4 substrate. Until more is known, use African wild potato cautiously or avoid in patients taking drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the concern with combining African Wild Potato and capmatinib (Tabrecta). Your body relies on a liver enzyme called CYP3A4 to clear capmatinib out of your system. In lab dish studies, this plant blocked that enzyme quite strongly. If the same thing happened in people, capmatinib could clear out more slowly and build up in your blood, which might mean more side effects like nausea, vomiting, swelling, tiredness, or changes in your liver or lung function.

I want to be honest about the evidence though: this is only test-tube data, and in one human study the plant did not change levels of a similar drug. So this is more of a theoretical caution than a proven problem. Since capmatinib is a cancer medicine, please loop in your oncologist or pharmacist before adding this supplement.

Tabrecta (capmatinib) is used to treat adults with metastatic non-small cell lung cancer (NSCLC) — lung cancer that has spread — whose tumors carry a specific genetic change called a MET exon 14 skipping mutation. A patient must test positive for this mutation using an FDA-approved test before starting treatment.

This is a targeted therapy, meaning it works against tumors with this particular genetic driver rather than all lung cancers.
