Why this applies to Loratadine

Loratadine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group African Wild Potato interacts with.

In vitro evidence shows that African wild potato extract can inhibit cytochrome P450 3A4 (CYP3A4) enzyme activity by up to 86%. Theoretically, African wild potato might increase levels of drugs metabolized by CYP3A4. However, in human research, African wild potato does not seem to affect the pharmacokinetics of Efavirenz, a CYP3A4 substrate. Until more is known, use African wild potato cautiously or avoid in patients taking drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the gist for Claritin. Your liver uses an enzyme called CYP3A4 (along with another one) to break down loratadine. In lab dishes, African Wild Potato has been shown to block CYP3A4 fairly strongly. On paper, that could mean a bit more loratadine hanging around in your body, which might bump up its side effects like drowsiness, dry mouth, or headache.

That said, I want to keep this in perspective. The concern comes only from test-tube studies, and in actual human research the potato didn't change levels of another similar drug. Loratadine is also a pretty gentle, non-drowsy antihistamine with a wide safety margin. So a serious problem is unlikely, but it's still smart to run this combo past your pharmacist or doctor first.
