Diphenhydramine falls under CNS DEPRESSANTS — the group Oregon Grape interacts with.
Animal research suggests that berberine, a constituent of Oregon grape, can have sedative effects.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the simple version: diphenhydramine (Benadryl) already makes most people drowsy. Oregon grape contains berberine, and in animal studies berberine showed mild sedative effects of its own. So the worry is that pairing the two could stack the sleepiness, leaving you groggier than you expected, plus dizzy, foggy-headed, or slow to react. That matters most if you're driving, working, or caring for others.
I'll be honest with you though, this is a theoretical concern based only on lab and animal data, not on real people. There's no human evidence showing a strong problem. Still, if you take Benadryl for sleep or allergies and also want Oregon grape, mention it to your pharmacist first, and watch how you feel before doing anything that needs you sharp.

Diphenhydramine temporarily relieves symptoms of hay fever, other upper respiratory allergies, and the common cold — including runny nose, sneezing, itchy or watery eyes, and itching of the nose or throat.
After Bite Xtra gel is formulated for topical use and is approved to temporarily relieve pain and itching from minor burns, minor cuts and scrapes, insect bites, minor skin irritations, and rashes from poison ivy, poison oak, and poison sumac.
