Why this applies to Zaleplon

Zaleplon falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Tangerine interacts with.

In vitro, tangeretin, a constituent of tangerine, induces a 52% increase in the metabolism of midazolam by cytochrome P450 3A4 (CYP3A4). This suggests that tangeretin may stimulate CYP3A4 activity. However, in humans, drinking tangerine juice 200 mL slightly delayed the absorption, but did not affect the metabolism, of midazolam, a CYP3A4 substrate. Theoretically, tangerine juice might increase CYP3A4 activity and decrease levels of drugs metabolized by this enzyme. However, this effect is unlikely.
Some drugs metabolized by CYP3A4 include amitriptyline (Elavil), amiodarone (Cordarone), citalopram (Celexa), felodipine (Plendil), lansoprazole (Prevacid), ondansetron (Zofran), prednisone (Deltasone, Orasone), sertraline (Zoloft), sibutramine (Meridia), and many others.

Here's the gist for your sleep aid: zaleplon (Sonata) is partly broken down by an enzyme called CYP3A4, and a compound in tangerine (tangeretin) was shown in lab studies to nudge that enzyme to work a little faster. In theory, that could clear zaleplon slightly quicker and make it a touch less effective, meaning you might not fall asleep as easily.

But honestly, I wouldn't lose sleep over this one. When researchers actually tested tangerine juice in people with a similar drug, it didn't really change how the drug was processed. The interaction is rated minor and unlikely, and zaleplon isn't a prodrug, so it doesn't flip the direction. Enjoy your tangerines, and if your sleep aid suddenly stops working well, just mention it to your pharmacist or doctor.

Zaleplon capsules are approved for the short-term treatment of insomnia. Specifically, they help reduce the time it takes to fall asleep (sleep onset). Clinical studies showed this benefit for up to 30 days of use.

Zaleplon has not been shown to increase total sleep time or reduce the number of times you wake up during the night. It is intended for short-term use, and if sleep problems continue beyond 7–10 days, a doctor should evaluate you for an underlying cause.
