Zolpidem falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Vitamin E interacts with.

Theoretically, vitamin E might induce metabolism of CYP3A4, possibly reducing the levels CYP3A4 substrates.
Vitamin E appears to bind with the nuclear receptor, pregnane X receptor (PXR), which results in increased expression of CYP3A4. Although the clinical significance of this is not known, use caution when considering concomitant use of vitamin E and other drugs affected by these enzymes.

Here's the gist for zolpidem (Ambien). Your body clears this sleep aid mainly through a liver enzyme called CYP3A4. There's a theory that high-dose vitamin E can nudge that enzyme to work a little faster. If that happens, your body could break zolpidem down quicker, which might leave you with a weaker effect, meaning you may not fall asleep as easily or stay asleep as well.

I want to be honest with you: this is only a theoretical concern based on how vitamin E behaves in the lab, not something we've clearly seen play out in real patients. Everyday food amounts of vitamin E shouldn't be an issue. If you take high-dose supplements and notice your sleep changing, just check in with your pharmacist or doctor before adjusting anything.

What Is Zolpidem Used For?

Brand names: Ambien CR, Intermezzo, Sublinox

Zolpidem is used short-term for insomnia. Most formulations — including Ambien, oral tablets, and Zolpidem Tartrate Capsules — are approved to help people who have trouble falling asleep at bedtime. Zolpidem Tartrate Capsules are specifically indicated for adults under 65.

The extended-release formulations (including Zolpidem Tartrate Extended-Release Tablets) help with both falling asleep and staying asleep through the night. Sublingual tablets (Edluar, Zolpidem Tartrate Sublingual Tablet, and Zolpidem tartrate sublingual) are taken as needed when you wake up in the middle of the night and cannot get back to sleep — but only if at least 4 hours of sleep time remain before you need to be up.
