Our pharmacy team’s full take, with four database checks built into the cards below — a summary of what is known, not a grade of the product itself.
What’s inside
Full disclosure
Pregnenolone 100 contains a single active ingredient: pregnenolone, a hormone precursor your body normally makes in small amounts. It's the raw material your system can convert into other hormones like estrogen, progesterone, testosterone, and DHEA, depending on what your body needs.
The tablet also contains several inactive ingredients—microcrystalline cellulose, vegetable oil, vegetable wax, magnesium stearate, vitamin E, and others—that help form and preserve the tablet.
Does it work?
Couldn't assess
We don't have established evidence in our data that pregnenolone is effective for any of the conditions listed—including Alzheimer's disease, autism spectrum disorder, back pain, bipolar disorder, or alcohol use disorder. All of these are rated as having insufficient reliable evidence.
If you're considering this supplement for a specific health concern, talk it over with your doctor.
How safe is it?
Well-documented data
Pregnenolone is generally well tolerated when taken by mouth, but it's a hormone with limited human safety data, so use it only under guidance from a healthcare professional. The most common side effects reported in studies include acne, agitation, drowsiness, diarrhea, sweating, tremor, and hair loss.
A few people have reported skin rashes, possibly related to sun sensitivity. Avoid pregnenolone during pregnancy and while breastfeeding—the safety data advises against it in both cases.
Meds to double-check
Moderate interaction found
Check with your doctor or pharmacist before taking this product if you use estrogens (including birth control pills or patches), progestins, testosterone therapy, progesterone, or benzodiazepines (anti-anxiety or sleep medications like diazepam). Pregnenolone may enhance the effects of hormone therapies or reduce how well anti-anxiety medications work.
The bottom line
Scorecard at a glanceFully disclosed formula with no assessable stated purpose. Moderate medication interactions have been identified, and safety information is well characterized.
Pregnenolone 100 is a hormone supplement that interacts with several common medication classes, especially hormonal contraceptives, hormone replacement therapy, anti-anxiety drugs, and testosterone or progesterone products. Pregnenolone 100 by NutriCology Drug Interactions
HelloPharmacist Interaction Report
Pregnenolone 100 by NutriCology interacts with several medication types through its single active ingredient, pregnenolone, which is a hormone precursor.
The most serious concern is with estrogens, progestins, testosterone, and progesterone—pregnenolone may theoretically enhance how these hormones work in your body, since it can convert into them. These are all Moderate-severity interactions.
Read the full breakdown — every affected drug type, severity by severity
Pregnenolone also interacts with benzodiazepines (the sedative class, like diazepam)—early research suggests it may reduce their calming effects when taken over time, possibly by changing how GABA receptors in your brain respond. This is also Moderate severity.
Altogether, these interactions span 82 individual medications.
Because pregnenolone is a hormone with limited human safety data, check your exact medications—especially any birth control, hormone replacement therapy, anti-anxiety medications, or testosterone or progesterone products—with your doctor or pharmacist before you start.

Pregnenolone
Benzodiazepines
Concomitant use of pregnenolone may reduce the effects of benzodiazepines.
Very preliminary clinical research shows that chronic use of pregnenolone reduces sedative effects of diazepam when compared with chronic use of placebo. This effect may be related to the activity of pregnenolone at GABAA receptors.
Estrogens
Theoretically, taking pregnenolone might enhance the effects of estrogens.
Pregnenolone is a precursor for several steroid hormones, including estrogens.
Progesterone
Theoretically, taking pregnenolone might enhance the effects of progesterone.
In humans, some research shows that oral administration of pregnenolone can increase levels of progesterone. However, other research shows no effect on progesterone levels. It is possible the impact of pregnenolone on progesterone levels may be dose dependent.
Progestin
Theoretically, taking pregnenolone might enhance the effects of progestin.
Pregnenolone is a precursor for several steroid hormones, including progestin.
Testosterone
Theoretically, taking pregnenolone might enhance the effects of testosterone.
Pregnenolone is a precursor for several steroid hormones, including testosterone. However, preliminary clinical research shows that taking pregnenolone 30-500 mg orally daily for up to 8 weeks does not affect testosterone levels.
