Why this applies to Pazopanib

Pazopanib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group African Wild Potato interacts with.

In vitro evidence shows that African wild potato extract can inhibit cytochrome P450 3A4 (CYP3A4) enzyme activity by up to 86%. Theoretically, African wild potato might increase levels of drugs metabolized by CYP3A4. However, in human research, African wild potato does not seem to affect the pharmacokinetics of Efavirenz, a CYP3A4 substrate. Until more is known, use African wild potato cautiously or avoid in patients taking drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the basic idea with Pazopanib (Votrient). Your liver uses an enzyme called CYP3A4 to clear this cancer drug from your body. In test-tube studies, African Wild Potato blocked that same enzyme quite strongly. If that played out in real life, your body might clear pazopanib more slowly, letting levels climb. That could mean more of its known side effects, such as high blood pressure, diarrhea, fatigue, liver irritation, or changes in your heart's rhythm.

That said, this concern comes only from lab work, not from people, and in one human study the supplement didn't affect a similar drug. So I wouldn't panic. Still, with a serious cancer medication like this, please don't add the supplement without first checking with your oncologist or pharmacist.

What Is Pazopanib Used For?

Brand name: Votrient

Pazopanib tablets — sold as the brand Votrient and as generic pazopanib — are used to treat two types of advanced cancer in adults: advanced renal cell carcinoma (RCC), the most common form of kidney cancer, and advanced soft tissue sarcoma (STS) in patients who have already received prior chemotherapy.

Important limitation: Pazopanib has not been shown to work in adipocytic soft tissue sarcoma (a subtype involving fat cells) or in gastrointestinal stromal tumors (GISTs), so it is not used for those cancers.
