Here's the simple version: Amantadine has some mild anticholinergic activity, meaning it gently dampens a nerve signal called acetylcholine. Calabar bean contains physostigmine, which does the opposite. It boosts acetylcholine. So in theory, the two push against each other, and the bean could slightly blunt the anticholinergic side of how amantadine works. For someone using amantadine for Parkinson's symptoms or drug-related movement problems, that might mean a touch less benefit.

I want to be honest, though: this is a theoretical concern based on how the chemicals behave, not on real cases, and it's rated minor and unlikely. The bigger issue is that Calabar bean itself is genuinely poisonous and is not a safe supplement to take at all. Please skip it and check in with your pharmacist or doctor about any concerns.

It is not a safe dietary supplement, and eating the bean can cause serious poisoning or death. Only the purified, regulated drug physostigmine has a legitimate medical use, and that must be prescribed and monitored by a doctor.

What Is Amantadine Used For?

Brand names: Symmetrel, Gocovri, Osmolex ER

Amantadine capsules, tablets, and oral solution are used to prevent or treat uncomplicated respiratory illness caused by influenza A virus strains. They are also used in the treatment of parkinsonism and drug-induced extrapyramidal reactions (movement problems caused by certain medications).

Gocovri (an extended-release amantadine capsule) has two specific approved uses: treating dyskinesia (uncontrolled, involuntary movements) in people with Parkinson's disease who are taking levodopa-based therapy, and as an add-on treatment to levodopa/carbidopa for people experiencing "off" episodes (periods when Parkinson's symptoms return or worsen).
