Animal research shows that calamus is a CNS depressant and increases gamma-aminobutyric acid levels.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Donnatal (belladonna alkaloids plus phenobarbital): the phenobarbital piece is a barbiturate that already calms your nervous system. Calamus appears to do something similar. In animal studies it acts like a mild brain depressant and seems to boost GABA, the same calming signal phenobarbital leans on. Put them together and the slowing effects could stack up.
In real life that might mean extra drowsiness, grogginess, dizziness, or feeling foggy and unsteady on your feet. I'll be honest, this is theoretical and based on lab and animal work, not strong human proof, so I'm not trying to scare you. Still, calamus carries its own safety baggage (the FDA pulled it from foods), so I'd skip it and check with your pharmacist or doctor first.

ANTICHOLINERGIC DRUGS

In vitro evidence shows that calamus can inhibit acetylcholinesterase (AChE).
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the idea behind this one. Products like Donnatal contain belladonna alkaloids, which work by blocking acetylcholine in your body to calm cramping and ease an upset stomach. Calamus appears to do the opposite in lab studies: it can block the enzyme that breaks down acetylcholine, so more of that messenger sticks around. In theory, that could push against the belladonna part and make it work less well, so you might notice more stomach cramping, looser stools, more watering eyes, or more saliva than you'd expect.
I want to be honest that this is only test-tube and animal evidence, not proven in people, so I wouldn't lose sleep over it. Still, calamus carries its own safety worries and is best avoided. Please check with your pharmacist or doctor before combining them.

Calamus is a swamp plant with a long history in Ayurvedic and traditional Chinese medicine, mostly for digestive and nervous-system complaints. However, it contains beta-asarone, a compound linked to cancer and other toxic effects in animal studies, which is why the U.S. FDA banned it from foods. Because of these safety concerns and a lack of solid human evidence, it is generally best avoided.
