Acetaminophen, Diphenhydramine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Celery interacts with.
In vitro and animal research suggests that constituents of celery can inhibit CYP1A2. This effect has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the situation with Tylenol PM, which pairs acetaminophen (a pain reliever) with diphenhydramine (the same antihistamine in Benadryl). Lab and animal studies hint that compounds in celery might slow down a liver enzyme called CYP1A2. If that enzyme runs slower, drugs it handles could clear a little less easily and build up slightly, which in theory could mean a bit more of the drowsiness, dry mouth, or grogginess that diphenhydramine already causes.
The honest truth: this is only a theoretical concern. It has never actually been shown in people, and eating celery as food is very unlikely to matter. Concentrated celery seed extracts are a different story. If you take those regularly, just mention it to your pharmacist or doctor.

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Tylenol PM (acetaminophen plus diphenhydramine) and celery. This isn't about your liver breaking the medicine down differently or the drug building up in your body. Instead, celery naturally carries light-sensitizing compounds called psoralens, and some medicines can also make your skin more touchy to sunlight. Put them together and, in theory, you might sunburn a little more easily.
Honestly, this one is very mild and based only on scattered, anecdotal reports, so I wouldn't lose sleep over it. Eating celery as food is unlikely to matter much. If you take concentrated celery seed supplements, just be a bit sun-smart: use sunscreen and cover up. Any questions, your pharmacist is happy to help.

ACETAMINOPHEN (Tylenol, others)
3 of 3Animal research suggests that concomitant use of celery juice plus acetaminophen prolongs the effects of acetaminophen. This effect has been attributed to a decrease in hepatic cytochrome P450 activity. However, other animal research shows that pretreatment with celery root extract protects against acetaminophen-induced acute liver failure. These effects have not been reported in humans.

Tylenol PM combines acetaminophen (the pain and fever reliever) with diphenhydramine (the same drowsy ingredient found in Benadryl). The interaction on file deals with the acetaminophen half. In some animal studies, celery juice appeared to slow down liver enzymes that break acetaminophen down, which could let it linger a bit longer in the body. Interestingly, other animal research suggested celery root might actually protect the liver. Bottom line: the science is mixed and has not been shown in people.
So I would not lose sleep over this one. Eating celery as food is a non-issue, and even concentrated celery supplements only theoretically nudge acetaminophen's effect. If you take this combo often, just stay within the labeled acetaminophen dose and check with your pharmacist.
