Here's the simple version. Coffee charcoal acts a bit like activated charcoal in your gut. Its porous surface can grab onto things passing through, and that includes pills you swallow. With Lonafarnib (Zokinvy), the worry is that taking the two close together could let the charcoal soak up some of your dose before your body absorbs it.

Lonafarnib is an active medicine, not a prodrug, so absorbing less of it simply means less drug working in your body, which matters for a treatment used in serious conditions like progeria. The good news is this is theoretical, based on how charcoal behaves rather than firm studies. If you use coffee charcoal, separating it from your Lonafarnib by at least two hours is the easy fix. Please run it by your pharmacist or doctor first.

About Lonafarnib

What Is Lonafarnib Used For?

Brand name: Zokinvy

Zokinvy (lonafarnib) is approved for patients 12 months of age and older who have a body surface area of 0.39 m² or above. It is used to reduce the risk of mortality in Hutchinson-Gilford Progeria Syndrome (HGPS) — a rare, severe premature-aging disorder.

Zokinvy is also used to treat processing-deficient Progeroid Laminopathies — a related group of rare genetic conditions — specifically those caused by a heterozygous LMNA mutation with progerin-like protein buildup, or by homozygous or compound heterozygous ZMPSTE24 mutations. It is not indicated for other Progeroid Syndromes or processing-proficient Progeroid Laminopathies.
