Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS
Why this applies to Darunavir, Cobicistat
Darunavir, Cobicistat falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Ergot interacts with.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the key thing with Prezcobix: it contains cobicistat, whose whole job is to block the liver enzyme CYP3A4 so your HIV medicine stays in your system longer. That same enzyme is what your body uses to clear ergot. So when you pair the two, the ergot can pile up instead of being broken down.
With ergot, more in the body is a real problem. It powerfully tightens blood vessels, and too much can cause cold, painful, numb hands and feet, severe headache, or dangerous circulation trouble (called ergotism). Because of this, ergot drugs are generally considered off-limits with strong CYP3A4 blockers like cobicistat. The evidence here is largely theoretical, but the risk is serious enough that I'd never combine them on your own. Please check with your pharmacist or doctor first.

About Ergot
What is Ergot?
- Heavy menstrual or uterine bleeding
- Bleeding after childbirth
- Migraine headaches
- Stimulating uterine contractions
Ergot is a toxic fungus that grows on rye and contains powerful chemicals (ergot alkaloids) that strongly narrow blood vessels and squeeze the uterus. Because raw ergot is poisonous and the dose is impossible to control safely, it should never be used as a home remedy or supplement. Some purified ergot chemicals are used as prescription drugs under strict medical supervision, but the crude herb is not safe to take.
