Repotrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) INDUCERS — the group Gravel Root interacts with.

Hepatotoxic pyrrolizidine alkaloids (PA) are substrates of cytochrome P450 3A4 (CYP3A4). Theoretically, drugs that induce CYP3A4 might increase the conversion of PAs to toxic metabolites. Some drugs that induce CYP3A4 include carbamazepine (Tegretol), phenobarbital, phenytoin (Dilantin), rifampin, rifabutin (Mycobutin), and others.

Here's the piece that matters with this combo: Gravel Root can contain compounds called pyrrolizidine alkaloids, and those are hard on the liver. Your body uses the enzyme CYP3A4 to turn them into their most toxic forms. Repotrectinib (Augtyro) is a mild CYP3A4 inducer, meaning it can speed that enzyme up. In theory, that could push more of Gravel Root's compounds into harmful liver-damaging byproducts.

I want to be honest about the evidence here: this concern comes from lab and animal work, not real patient studies, so it's more of a caution than a proven danger. Still, Gravel Root already has liver-safety worries on its own, and cancer therapy is not something to add risk to. Please check with your oncologist or pharmacist before using Gravel Root while on Repotrectinib.
