Glutethimide falls under CYTOCHROME P450 3A4 (CYP3A4) INDUCERS — the group Gravel Root interacts with.

Hepatotoxic pyrrolizidine alkaloids (PA) are substrates of cytochrome P450 3A4 (CYP3A4). Theoretically, drugs that induce CYP3A4 might increase the conversion of PAs to toxic metabolites. Some drugs that induce CYP3A4 include carbamazepine (Tegretol), phenobarbital, phenytoin (Dilantin), rifampin, rifabutin (Mycobutin), and others.

Here's the concern with pairing Gravel Root and Glutethimide (Doriden). Gravel Root contains compounds called pyrrolizidine alkaloids that can be hard on the liver. Your body uses an enzyme called CYP3A4 to process these compounds, and Glutethimide is one of the medications that can speed that enzyme up. In theory, that faster processing could create more of the liver-damaging byproducts from the herb, raising the risk of liver harm.

I want to be honest about the evidence here: this is based on lab and animal studies, not real-world human cases, so it's more of a theoretical worry. Still, since Gravel Root already has questionable safety and weak proof it even works, I'd skip it and talk with your doctor or pharmacist before combining the two.
