Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Guggul interacts with.

Theoretically, guggul might reduce the effects of CYP3A4 substrates.
In vitro research shows that guggul constituents known as guggulsterones can induce CYP3A4.

Here's the concern with pairing guggul and entrectinib. Your body clears entrectinib using a liver enzyme called CYP3A4. Lab studies suggest the active parts of guggul (guggulsterones) may rev up that enzyme. If that happens in people, your body could clear entrectinib faster than it should, leaving less of it in your bloodstream to do its job.

For a cancer medicine like this, that matters. Lower levels could mean the drug works less well against the tumor, and that's not a risk worth taking lightly. Keep in mind this is based only on lab research, not human studies, so it isn't proven. Still, with a serious medication like entrectinib, please check with your oncologist or pharmacist before using guggul.

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek (entrectinib) is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC), as confirmed by an FDA-approved test. It is also approved for adult and pediatric patients older than 1 month of age who have solid tumors — any cancer type — that carry an NTRK gene fusion detected by an FDA-approved test, provided the tumor has no known acquired resistance mutation, is metastatic or cannot be surgically removed without severe harm, and has progressed after prior treatment or has no satisfactory alternative therapy.

The NTRK gene fusion approval in solid tumors was granted under accelerated approval, meaning it is based on tumor response rates. Continued approval may depend on results from confirmatory clinical trials.
