Why this applies to Rucaparib Camsylate

Rucaparib Camsylate falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Valerian interacts with.

Valerian does not seem to have a clinically relevant effect on levels of drugs metabolized by CYP2D6.
Although some in vitro evidence suggests that valerian affects CYP2D6, clinical pharmacokinetic (PK) studies show that valerian is unlikely to affect the CYP2D6 enzyme. In one PK study, taking valerian 1000 mg (providing about 11 mg valerenic acid) nightly for 14 days did not affect the metabolism of dextromethorphan, a CYP2D6 substrate. In another PK study, taking valerian 125 mg three times daily for 28 days did not affect metabolism of debrisoquine, an accepted CYP2D6 probe-substrate.

Here's the good news on pairing valerian with your Rubraca (rucaparib). On paper, rucaparib is handled in part by a liver enzyme called CYP2D6, and some early lab studies hinted that valerian might tinker with that enzyme. But when researchers actually tested this in people, valerian didn't meaningfully change how CYP2D6 cleared other medications. So we don't expect valerian to push your rucaparib levels up or down in a way that matters.

That said, rucaparib already carries side effects like nausea, fatigue, and low blood counts, and valerian can make you drowsy on its own. I'd still run this combination past your oncology team before adding it, just so everyone is on the same page with your cancer treatment.

About Rucaparib

What Is Rucaparib Used For?

Brand name: Rubraca

Rubraca (rucaparib) is used for two types of cancer in adults who carry a harmful BRCA gene mutation (either inherited or found in the tumor). First, it is used as maintenance treatment — meaning ongoing treatment to keep cancer from coming back — in people with recurrent epithelial ovarian cancer, fallopian tube cancer, or primary peritoneal cancer who responded (fully or partially) to platinum-based chemotherapy.

Second, Rubraca treats metastatic castration-resistant prostate cancer (advanced prostate cancer that has stopped responding to hormone-lowering treatment) in adults who have already received androgen receptor-directed therapy. Patients must be tested with an FDA-approved companion diagnostic to confirm the BRCA mutation before starting Rubraca for prostate cancer.
