Insufficient evidence
The evidence for this product's active ingredients is mixed and incomplete. L-phenylalanine is possibly effective for vitiligo but possibly ineffective for ADHD, with insufficient evidence for alcohol use disorder and multiple sclerosis.
L-Histidine shows possibly effective evidence only for cholera; the data is insufficient for eczema, kidney failure, and rheumatoid arthritis. L-Lysine is possibly effective for cold sores, but evidence is insufficient for child growth, pressure ulcers, and diabetic foot ulcers.
L-Threonine appears possibly ineffective for ALS and has insufficient evidence for multiple sclerosis, spasticity, and related conditions. We hold no effectiveness data for L-leucine, L-isoleucine, L-valine, or the proprietary blends in this formula.
How safe is it?
Well-documented data
L-Phenylalanine is generally well tolerated at typical doses, but the safety data advises against supplement doses during pregnancy and breastfeeding. Common side effects include anxiety, constipation, headache, heartburn, insomnia, nausea, and sedation.
Anyone with PKU (phenylketonuria), a genetic condition, should avoid this supplement. L-Histidine has limited safety data at high doses; the data advises caution in pregnancy and with supplemental use.
L-Threonine from food is safe, but supplement safety is not well studied, especially in pregnancy and breastfeeding — the data advises caution. Reported side effects include headache and minor gastrointestinal upset like diarrhea, gas, and constipation.
L-Lysine is generally well tolerated, though high amounts may cause stomach upset; long-term safety is not well studied. Pregnancy and breastfeeding safety for L-lysine supplements is not well established — talk to your doctor first.

Jolly Watermelon by Purus Labs contains several amino acids, and we've found documented interactions with a number of medications. The most serious concern is L-phenylalanine, which can significantly reduce the effectiveness of levodopa (a medication for Parkinson disease) by competing with it for transport into the brain — this can worsen tremor, stiffness, and the "on-off" effect.
L-phenylalanine also moderately reduces how your body absorbs baclofen (a muscle relaxant) and may theoretically raise blood pressure risk if you take an older class of antidepressants called MAOIs.

L-Phenylalanine
Levodopa
Phenylalanine, especially in high doses, can reduce the effectiveness of levodopa.
Phenylalanine competes with levodopa for carrier-mediated transport into the brain. The resulting reduction in levels of levodopa in the brain can exacerbate tremor, rigidity, and the "on-off" phenomenon in patients with Parkinson disease.
Baclofen
Concomitant intake of phenylalanine may reduce the intestinal absorption of baclofen.
Phenylalanine and baclofen share the same intestinal carrier for absorption; phenylalanine competitively inhibits the absorption of baclofen, reducing its plasma levels.
Monoamine Oxidase Inhibitors (Maois)
Theoretically, concomitant use of L-phenylalanine and non-selective MAOIs might increase the risk of hypertensive crisis.
L-phenylalanine is metabolized to tyrosine. Some evidence suggests that L-phenylalanine, given with the non-selective MAOI pargyline, might prevent the elimination of tyramine, increasing the risk of hypertensive crisis. However, this was not reported in a small number of patients when using L-phenylalanine with the partially selective MAO-B inhibitor, selegiline.
L-Threonine
Nmda Antagonists
Theoretically, threonine might decrease the effects of NMDA antagonists.
Threonine increases central nervous system (CNS) glycine levels. Glycine seems to bind a site on NMDA receptors and enhance the activity of the receptors.
L-Lysine
5-Ht4 Agonists
Theoretically, lysine may reduce the effects of 5-HT4 agonists.
Animal research suggests that L-lysine is a partial serotonin receptor 4 (5-HT4) antagonist and inhibits diarrhea induced by the 5-HT4 agonist, 5-hydroxytryptophane.
